摘 要:目的 检测微小RNA-145(miR-145)对兔快速心房起搏模型心房有效不应期(AERP)、房颤诱发率(AFIR)及心房电重构(AER)的影响。方法 80只新西兰大耳白兔随机分成假手术组、模型组、激动剂组和激动剂对照组(n=20)。假手术组不予快速心房起搏,模型组持续快速心房起搏3周,激动剂组和激动剂对照组分别于起搏前1周通过耳缘静脉注射miR-145agomir和agomir control,观察起搏后AERP和AFIR。采用实时荧光定量PCR检测各组心房肌组织中miR-145表达,采用蛋白质印记检测肌浆网钙泵(ATP2A2)和L型Ca2+通道α1C亚基(CACNA1C)及兰尼碱受体2(RYR2)表达。结果 模型组miR-145表达低于假手术组,激动剂组miR-145表达高于模型组与激动剂对照组(P<0.05)。模型组起搏后AERP较假手术组缩短,激动剂组起搏后AERP较模型组和激动剂对照组延长(P<0.05)。模型组AFIR高于假手术组,激动剂组AFIR低于模型组和激动剂对照组(P<0.05)。模型组CACNA1C和ATP2A2 mRNA与蛋白表达较假手术组下调,而RYR2 mRNA与蛋白表达上调(P<0.05)。激动剂组CACNA1C和ATP2A2 mRNA与蛋白表达较模型组和激动剂对照组上调,而RYR2 mRNA与蛋白表达下调(P<0.05)。结论 超表达miR-145延长新西兰大耳白兔快速心房起搏模型AERP并降低AFIR,其机制可能与增加CACNA1C和ATP2A2表达并降低RYR2表达而影响AER有关。Objective To observe the effects of microRNA-145(miR-145) on the atrial effective refractory period(AERP),atrial fibrillation induction rate(AFIR) and atrial electrical remodeling(AER) in a rabbit rapid atrial pacing model.Methods 80 New Zealand big-eared white rabbits were randomly divided into sham operation,model,agonist and agonist control groups(n =20),respectively.The sham operation group was not given rapid atrial pacing,the model group was continuously given rapid atrial pacing for 3 weeks,the agonist group and the agonist control group were injected with miR-145 agomir and agomir control through the ear vein at 1 week before pacing,and the AERP after pacing and AFIR were observed.Real-time fluorescent quantitative PCR was used to detect the expression of miR-145 in the atrial muscle tissues of each group,and the expression of sarcoplasmic reticulum calcium ATPase(ATP2A2),L-type Ca2+ channel alpha 1C subunit(CACNA1C) and ryanodine receptor type 2(RYR2) was detected by western blotting.Results The expression of miR-145 in the model group was lower than that in the sham operation group,and the expression of miR-145 in the agonist group was higher than that in the model group and the agonist control group(P <0.05).The AERP after pacing in the model group was shorter than that in the sham operation group,and the AERP after pacing in the agonist group was longer than that in the model group and the agonist control group(P <0.05).The AFIR in the model group was higher than that of the sham operation group,and the AFIR in the agonist group was lower than that of the model group and the agonist control group(P <0.05).The expression of CACNA1C and ATP2A2 mRNA and protein in the model group was downregulated compared with the sham operation group,while the expression of RYR2 mRNA and protein was upregulated(P<0.05).The expression of CACNA1C and ATP2A2 mRNA and protein in the agonist group was upregulated compared with the model group and the agonist control group,while the expression of RYR2 mRNA and protein was do
