蒙药蓝刺头对去卵巢大鼠BMD、E_(2)及股骨头CYP450的作用

摘　　要：目的:通过研究蒙药蓝刺头对去卵巢大鼠骨密度(bone mineral density,BMD)、血清雌二醇(estradiol2,E_(2))、股骨头中芳香化酶P450(aromatase cytochrome P450,CYP450)的作用,探讨蓝刺头对绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)的作用机制。方法:取Wistar雌性大鼠105只,随机分为己烯雌酚组,强骨胶囊组,蒙药蓝刺头低、中、高剂量组,假手术组,模型组7组,每组15只,复制去卵巢大鼠骨质疏松模型,行阴道上皮角化实验,确定造模成功后,在正常条件下饲养90天,灌胃90天后取血,采用酶联免疫法测血清中E_(2),分离左侧股骨双能X线测BMD,免疫组织化学法测大鼠右侧股骨头CYP450表达。结果:蒙药蓝刺头中剂量组可提高去卵巢大鼠BMD、血清E_(2)水平,增强股骨头CYP450表达。结论:蒙药蓝刺头可促进雌激素转化酶、前体物质表达,对治疗去卵巢大鼠骨质疏松症具有明显作用,为进一步防治PMOP,深入阐释中医“肾主骨”理论提供了实验依据。Objective:To survey the mechanism of Lancitou in treating postmenopausal osteoporosis(PMOP)by studying mongolian medicine Lancitou on bone mineral density(BMD),serum estradiol2(E_(2))and aromatase cytochrome P450(CYP450)in femoral bone of ovariectomized rats.Methods:All 105 Wistar female rats were selected and randomized into diethylstilbestrol(DES)group,Qianggu capsules group,high,moderate and low doses groups of Lancitou,sham operation group and the model group,15 rats each group.PMOP models were duplicated to perform vaginal cornification test,after confirming successful modeling,the rats were raised for 90 days under normal conditions,the blood was taken after drenching for 90 days,ELISA method was adopted to detect E_(2),left femur dual energy X-ray was separated to measure BMD,immunohistochemical method to detect the expressions of CYP450 in right caput femoris.Results:Mongolian medicine Lancitou of moderate dose could lift BMD and E_(2) expressions,enhance CYP450 expressions.Conclusion:Mongolian medicine Lancitou could lift BMD and E_(2) expressions,enhance CYP450 expressions,it is proved that it could promote the expressions of estrogen invertase and precursor substances,it possesses notable effects of treating osteoporosis of ovariectomized rats,and it could provide experimental reference for further prevention and treatment for PMOP,deeply illustration of TCM"kidney governing bone"theory.
