摘  要:牡丹具有产油率高,富含亚麻酸、亚油酸和油酸等超长链不饱和脂肪酸含量高等特点,是一种优质的木本油料作物.然而,牡丹基因组中KCS基因的存在情况及其演化特征鲜有报道.从牡丹基因组中鉴定出13条KCS基因,蛋白质的氨基酸序列长度在274~967aa之间,它们编码的蛋白质中均含有3-Oxoacyl-[acyl-carrier-protein(ACP)]synthaseⅢC结构域和TypeⅢpolyketide synthase-like结构域.系统发育分析显示,牡丹KCS基因组成4个演化分支.同一演化分支或近缘演化分支中的KCS基因编码蛋白质的结构域和氨基酸基序相似,而演化关系较远演化分支中的基因结构差异较大.研究牡丹KCS基因的演化特征能进一步发掘牡丹KCS基因的功能,加深对其超长链脂肪酸合成机制的了解,为油用牡丹的分子遗传育种提供一定的理论指导.Tree peony is a kind of high quality woody oil crop with high oil yield and high content of ultra-long chain unsaturated fatty acids such as linolenic acid,linoleic acid and oleic acid.However,studies on the KCS genes in the tree peony genome have not been performed.In this study,13 KCS genes were identified in the genome of tree peony,and the amino acid sequence length of coding proteins ranged from 274 aa to 967 aa.All of these proteins have two relatively conserved structure,including 3-Oxoacyl-[acyl-carrier-protein(ACP)]synthaseⅢC domain and TypeⅢpolyketide synthase-like domain.The phylogenetic analysis revealed that the KCS genes formed four monophyletic clades.The protein domains and motifs encoded by KCS genes within the same or closely related clades were similar,but was different between the clades.Analysis of the KCS gene family in tree peony could further explore the KCS gene functions and the mechanism of fatty acid synthesis.Meanwhile,it could provide some theoretical guidance for the molecular genetic breeding of tree peony.
