摘  要:目的观察桂枝加葛根汤对颈椎病模型兔不同节段颈椎纤维环细胞凋亡的影响。方法将24只6月龄新西兰兔随机分为空白组、模型组和桂枝加葛根汤组,每组8只。模型组和桂枝加葛根汤组新西兰兔使用颈椎固定装置制备颈椎病模型,造模2个月后,桂枝加葛根汤组予以桂枝加葛根汤浓缩液灌胃,空白组和模型组予以等体积生理盐水灌胃,干预2周后处死取材。采用苏木精-伊红(HE)染色观察纤维环组织形态学改变,实时荧光定量聚合酶链式反应(Real-time PCR)检测纤维环组织凋亡相关因子mRNA表达水平。结果与空白组相比,模型组的纤维环细胞数量变少、界限不清,而桂枝加葛根汤组的纤维环细胞数量较模型组多,界限清楚。在相同的组别中,C_(5-6)节段的纤维环细胞数量最少,组织形态不规整,其次是C_(6-7),最后是C_(4-5)。在C_(5-6)和C_(6-7)两个节段颈椎中,与空白组相比,模型组纤维环细胞BaxmRNA、Caspase-3mRNA表达上升,而Bcl-2mRNA表达下降(P<0.05);与模型组相比,桂枝加葛根汤组纤维环细胞BaxmRNA、Caspase-3mRNA表达下降,而Bcl-2mRNA表达上升(P<0.05);而在C_(4-5)节段颈椎中,BaxmRNA,Caspase-3 mRNA、Bcl-2mRNA的表达差异无显著差异(P>0.05)。结论桂枝加葛根汤可以维持纤维环的正常形态,抑制纤维环细胞的凋亡,对C_(5-6)节段的影响较显著,其次是C_(6-7),而对C_(4-5)节段的影响不太明显。Objective To observe the effect of Guizhi Plus Gegen Decoction(GGD)on apoptosis of annulus fibrosus cells in different segments of cervical vertebra of rabbits with cervical spondylosis.Methods Twenty-four 6-month-old rabbits were randomly divided into blank group,model group,GGD group.One month aftermodeling by self-made cervical spine fixation device,GGD group was given GGD by gavage,blank group and model group were given 0.9%saline by gavage,respectively.After 2 weeks of intervention,the rabbits were sacrificed fordetection.Morphological changes in annulus fibrosus was detected by HE staining,while the mRNA expressions of Bcl-2,Caspase-3 and Baxin annulus fibrosus tissue were detected by quantitative real-time polymerase chain reaction(Read-time PCR).Results Compared with blank group,the number of annulus fibrosus cells in model group was less and the boundary was unclear,while the number of annulus cells in GGD group was more than that in model group.In the same group,the number of annulus fibrosus cells in C_(5-6)segment was the least,and the degree of degeneration was the most serious,followed by C_(6-7),and finally C_(4-5).Compared with tblank group,the mRNA expression of Bax and Caspase-3 in the annulus fibrosus of model group increased,while the mRNA expression of Bcl-2 decreased(P<0.05).Compared with model group,the mRNA expression of Bax and Caspase-3 in the annulus fibrosus tissue of GGD group decreased,while the mRNA expression of Bcl-2 increased(P<0.05).The difference of this change was significant in C_(5-6)and C_(6-7)segments,but not significant in C_(4-5)segments.Conclusions GGD could maintain the normal shape of annulus fibrosus,and inhibit the mRNA expression of Bax and Caspase-3 in annulus fibrosus,and promote the mRNA expression of Bcl-2.The effect on C_(5-6)segment is more significant,followed by C_(6-7),but not on C_(4-5)segment.
