摘 要:目的对2020年版《中国药典》大黄游离蒽醌含量测定项下的提取方法进行改进,并通过正交试验法来优化大黄游离蒽醌的提取工艺。方法采用HPLC法测定大黄游离蒽醌含量,通过单因素考察比较回流提取与超声提取的提取量,并以料液比、提取时间、提取次数为因素采用正交试验法优选大黄游离蒽醌的最佳提取方案。结果回流1h与超声30min游离蒽醌的提取量没有显著性的差异(P>0.05),超声提取更为便捷,考察的最佳提取工艺条件为料液比0.1∶100,超声时间30min,提取1次。结论超声提取法更适合大黄游离蒽醌的提取,优化后的提取方法操作简便,耗时短,可信度高,测定结果重复性好。Objective The extraction method under the determination item was improved,and the extraction process of rhubarb free anthraquinone was optimized by orthogonal test method.Methods The content of free anthraquinone in rhubarb was determined by HPLC.Compare the extraction volume of reflux extraction and ultrasonic extraction through single factor investigation.And taking the ratio of material to liquid,extraction time,and extraction times as factors,an orthogonal test method was used to optimize the best extraction scheme for free anthraquinone from rhubarb.Results There is no significant difference in the extraction amount of free anthraquinone between reflux for 1h and ultrasonic for 30min(P>0.05),but ultrasonic extraction is more convenient.The optimal extraction process conditions investigated were 0.1∶100 material-to-liquid ratio,30min of ultrasonic time,and one extraction.Conclusion The ultrasonic extraction method is more suitable for the extraction of free anthraquinone from rhubarb.The optimized extraction method is simple to operate,time-consuming,high in reliability,and the measurement results are good in repeatability.
