摘  要:目的运用超高效液相色谱-串联质谱(UPLC-MS/MS)技术评估金荞麦对大鼠CYP450酶活性影响。方法24只大鼠随机分成金荞麦低剂量、中剂量、高剂量3组和对照组。金荞麦组给予大鼠金荞麦煎煮液(0.15 g/ml),低剂量组0.5 ml/只,中剂量组1.0 ml/只,高剂量组2.0 ml/只,对照组给予大鼠生理盐水2 ml/kg,持续给药30 d。30 d以后,4组灌胃给予大鼠5种探针药物,采集血浆样品,后用UPLC-MS/MS检测血浆中探针药及其代谢产物的浓度。结果与对照组对比,金荞麦的低剂量组的美托洛尔AUC、C_(max)升高(P<0.05),CL降低(P<0.05)。与对照组对比,金荞麦中剂量组和高剂量组的羟基美托洛尔C_(max)升高(P<0.01)。与对照组对比,金荞麦低剂量的羟基咪达唑仑AUC和C_(max)降低(P<0.05),CL升高(P<0.05),中剂量组的C_(max)降低(P<0.05)。结论金荞麦对大鼠CYP2B1、CYP1A2和CYP2C9酶活性影响不大。Objective To evaluate the effect of Fagopyrum dibotrys on CYP450 enzyme activity in rats by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Methods A total of 24 rats were randomly divided into high-dose,medium-dose and low-dose Fagopyrum dibotrys groups and control group.The Fagopyrum dibotrys group was given Fagopyrum dibotrys decoction(0.15 g/ml),0.5 ml for one rat in the low-dose group,1.0 ml for one rat in the middle-dose group,2.0 ml for one rat in the high-dose group,and the control group was given saline 2 ml for one rat,continues for30 days.Then,the four groups were given 5 probe drugs by intragastric administration,and plasma samples were collected.UPLC-MS/MS was used to detect the concentration of probe drugs and their metabolites in plasma.Results Compared with the control group,the AUC and C_(max)of metoprolol in the low-dose group of Fagopyrum dibotrys increased(P<0.05),while CL decreased(P<0.05).Compared with the control group,the C_(max)of hydroxymetoprolol increased in the middle-dose and high-dose groups of Fagopyrum dibotrys(P<0.01).Compared with the control group,AUC and C_(max)of low-dose hydroxymidazolam decreased(P<0.05),CL increased(P<0.05),and C_(max)of the middle-dose group decreased(P<0.05).Conclusion Fagopyrum dibotryshas little effect on the CYP2B1,CYP1A2 and CYP2C9enzyme activities in rats.
