摘 要:目的探讨宝鸡市人间布鲁菌病流行特征,并对分离株多位点序列分型基因分析,为布鲁菌病的预防和控制提供依据。方法收集宝鸡市2019年—2021年4月分离于临床的6株布鲁菌,采用VITEK 2 Compact及MALDI-TOF MS进行细菌种属鉴定,多位点可变数目串联重复序列分析(MLVA)进行分型,并与其他已报道菌株聚类分析。结果6株布鲁菌分离株检测结果与传统分型基本相符,为1个种(羊种)和2个生物型(羊2型和羊4型);MLVA-16将其分为6种基因型。与国内其他地区20株布鲁菌聚类分析显示,河南的2011Jiang#035菌株与宝鸡地区菌株的亲缘性最近,上海的2011Jiang#065与hong#003、hong#004亲缘性近。结论宝鸡地区流行的主要是羊种布鲁菌3型,种型和生物型较单一。MLVA-16是一种分子基因分型工具,能够证明疫情暴发和追溯调查的流行病学之间的联系,且有助于提高布鲁菌病控制方案的有效性。Objective Discuss the epidemiological characteristics of human brucellosis in Baoji City,and analyze the multi-locus sequence typing gene of the isolates to provide a basis for the prevention and control of brucellosis.Methods Six strains of Brucella isolated from Baoji City from April,2019 to 2021 were identified by VITEK 2 Compact and MALDI-TOF MS,and classified by MLVA.Cluster analysis was performed with other reported strains.Collect 6 strains of Brucella isolated from the clinic in Baoji City in 2019,use VITEK 2 Compact and MALDI-TOF MS for bacterial species identification,multilocus variable number tandem repeat analysis(MLVA)for typing,and compare them with others Cluster analysis of strains has been reported.Results The test results of 6 Brucella isolates are basically consistent with the traditional typing,which are 1 species(sheep species)and 2 biotypes(sheep type 3 and sheep type 4);MLVA-16 divides them into 6 genotypes.Cluster analysis with 20 strains of Brucella from other regions in China shows that the 2011Jiang#035 strain in Henan is closest to the Baoji strains,and the 2011Jiang#065 in Shanghai is closely related to hong#003 and hong#004.Conclusion The type 3 Brucella melitensis is prevalent in Baoji area,and the species and biotypes are relatively single.MLVA-16 is a molecular genotyping tool that can prove the epidemiology link between outbreaks and retrospective investigations,and helps improve the effectiveness of brucellosis control programs.
