摘 要:为了解决生物骨植入材料杂萘联苯聚芳醚酮(PPEK)生物活性较差的问题,采用化学方法对PPEK进行改性,得到了羟基化改性PPEK和磺化改性PPEK。通过溶液旋涂的方法把两种改性聚合物涂覆在PPEK薄片表面,分别得到表面羟基化的PPEK样品(PPEK-OH)和表面磺化的PPEK样品(PPEK-SO_(2)H)。用X射线光电子能谱和接触角测试仪表征了改性前后PPEK表面的元素组成和亲疏水性,同时,用耗散型石英晶体微天平对PPEK-OH和PPEK-SO_(2)H表面的蛋白吸附行为进行表征。结果表明,PPEK-OH样品和PPEK-SO_(2)H样品表面人体纤连蛋白吸附密度分别增加了20.5%和33.8%。用MC3T3-E1成骨细胞评价PPEK-OH和PPEK-SO_(2)H的细胞相容性和成骨分化活性,结果表明,与未改性PPEK相比,PPEK-OH样品和PPEK-SO_(2)H样品表面的细胞贴附与成骨活性得到显著改善。To solve the problem that the biological bone implant material poly(phthalazinone ether ketone)(PPEK)has the poor bioactivity,chemical modification approaches were used in PPEK to obtain hydroxylated PPEK and sulphonated PPEK.The two modified polymers were coated on the surface of PPEK flakes by solution spin coating to obtain surface hydroxylated PPEK samples(PPEK-OH)and surface sulphonated PPEK samples(PPEK-SO_(2)H),respectively.The elemental composition and hydrophobicity of the unmodified and modified PPEK surface were characterized by X-ray photoelectron spectroscopy and contact angle tester.The protein adsorption density of the PPEK-OH and PPEK-SO_(2)H surfaces were also characterized by quartz crystal microbalance with dissipative(QCM-D).The results show that the adsorption density of human fibronectin on the surfaces of PPEK-OH and PPEK-SO_(2)H samples increase by 20.5%and 33.8%,respectively.The cytocompatibility and osteogenic differentiation activity of PPEK-OH and PPEK-SO_(2)H were evaluated using MC3T3-E1 osteoblasts.The results show that the cytosolic adhesion and osteogenic activity on the surface of PPEK-OH and PPEK-SO_(2)H samples are significantly improved compared to unmodified PPEK.
