摘　　要：目的评估傅里叶红外光谱技术(FTIR)对金黄色葡萄球菌快速分型的能力。方法收集21株金黄色葡萄球菌,使用FTIR检测这些菌株在900/cm~1200/cm区域的红外吸收光谱,对菌株的同源性进行分析。同时对菌株进行全基因组测序(WGS),分析各菌株的多位点序列分型(MLST)、spa分型以及核心基因组的单核苷酸多态性(SNP)分型。并以WGS的SNP分型结果作为金标准,评估FTIR与其他分型方法对金黄色葡萄球菌的分型能力差异;同时对2种培养基(血平板和MH平板)的FTIR结果进行评估。结果分离自血平板的金黄色葡萄球菌的FTIR结果与WGS分型方法的一致性最好。FTIR的分辨率与WGS方法相当,高于MLST和spa分型方法。结论FTIR对金黄色葡萄球菌的区分能力具有较高准确性,且操作简便快速,大大缩短了分型结果的周转时间,同时成本低廉,具有良好的临床推广前景。Objective To evaluate the ability of Fourier transform-infrared spectroscopy(FTIR)for rapid typing of Staphylococcus aureus.Methods Twenty-one S.aureus strains were collected and identified using the FTIR by detecting the infrared absorption spectra of the strains in 900 cm-1200 cm.Meanwhile,the strains were subjected to whole-genome sequencing(WGS),and then,MLST,spatyping,and single nucleotide polymorphism(SNP)typing of the core genome of each strain were analyzed.The SNP typing results of WGS were used as the gold standard to evaluate the discrepancy in typing ability of FTIR and other typing methods for S.aureus strains.In addition,we evaluated the strain discernibility discrepancy between the blood agar plate and MH agar plate through FTIR.Results FTIR results of S.aureus isolated from the blood agar plate had the best consistency with the WGS typing method.The resolution of FTIR is comparable to that of the WGS typing method and is higher than that of MLST and spa typing methods.Conclusion FTIR has high accuracy in distinguishing S.aureus homology,together with the advantages such as simple to operate,low costs,and short turn-around time,which has a good clinical promotion prospect.
