不同剂型蒙药暖宫七味的镇痛药效学研究

摘　　要：目的:观察不同剂型蒙药暖宫七味对家兔离体肠平滑肌及小鼠醋酸所致疼痛的影响,并进行药效学比较。方法:将80只SPF级雌性KM小鼠随机分为空白组、阳性药组和暖宫七味散及暖宫七味胶囊低剂量组(0.78 g·kg^(-1))、中剂量组(1.56 g·kg^(-1))、高剂量组(2.18 g·kg^(-1)),每组10只。空白组和阳性药组小鼠给予同体积羧甲基纤维素钠,其余各组小鼠给予相应药物灌胃15 d,末次给药30 min后,分别采用热板法和醋酸扭体法考察暖宫七味胶囊对小鼠疼痛阈值时间及出现扭体反应时间的影响。将6只清洁级雌性日本大耳白兔随机分为暖宫七味散低剂量组、暖宫七味散高剂量组和暖宫七味胶囊低剂量组、暖宫七味胶囊高剂量组,采用BL-420F生物机能实验系统分析暖宫七味对家兔肠平滑肌收缩功能的影响。结果:离体兔肠实验表明,低剂量的暖宫七味散和暖宫七味胶囊可影响离体兔肠平滑肌的收缩功能。热板实验表明,与空白组比较,阳性药组和暖宫七味散中剂量组、暖宫七味散高剂量组及暖宫七味胶囊中剂量组、暖宫七味胶囊高剂量组小鼠的疼痛阈值时间延长(P<0.05);与阳性药组比较,暖宫七味散低剂量组、暖宫七味散中剂量组和暖宫七味胶囊低剂量组、暖宫七味散中剂量组小鼠的疼痛阈值时间缩短(P<0.05)。高剂量的暖宫七味散和暖宫七味胶囊对小鼠的疼痛阈值与阳性药组比较,差异无统计学意义(P>0.05)。醋酸扭体实验表明,与空白组比较,阳性药组和暖宫七味散低剂量组、暖宫七味散高剂量组及暖宫七味胶囊低剂量组、暖宫七味胶囊中剂量组、暖宫七味胶囊高剂量组小鼠扭体时间延长(P<0.05);与阳性药组比较,暖宫七味散低剂量组、暖宫七味散中剂量组和暖宫七味胶囊低剂量组、暖宫七味胶囊中剂量组、暖宫七味胶囊高剂量组小鼠扭体时间缩短(P<0.05),而暖宫七味散高剂量组小�Objective:To observe the effects of different dose forms of Mongolian medicine Seven-Ingredient Formulas for Warming Uterus on isolated intestinal smooth muscle of rabbits and pain caused by acetic acid in mice,and to compare its pharmacodynamics.Methods:A total of 80 SPF-class female KM mice were randomly divided into the blank group,the positive drug group,the low-dose group of Seven-Ingredient Powder for Warming Uterus(0.78 g·kg^(-1)),the medium-dose group of Seven-Ingredient Powder for Warming Uterus(1.56 g·kg^(-1)),the high-dose group of Seven-Ingredient Powder for Warming Uterus(2.18 g·kg^(-1)),the low-dose group of Seven-Ingredient Capsules for Warming Uterus(0.78 g·kg^(-1)),the medium-dose group of Seven-Ingredient Capsules for Warming Uterus(1.56 g·kg^(-1)),the high-dose group of Seven-Ingredient Capsules for Warming Uterus(2.18 g·kg^(-1)),with 10 mice in each group.The mice in the blank group and the positive control group were given the same volume of CMC Na,and the mice in the other groups were given corresponding drugs by gavage for 15 days.And 30 minutes after the last administration,the effects of Seven-Ingredient Formulas for Warming Uterus on the pain threshold time and body twisting reaction time of mice were investigated by hot plate method and acetic acid writhing method respectively.Six clean-grade female Japanese white rabbits were randomly divided into the low-dose group of Seven-Ingredient Powder for Warming Uterus,the high-dose group of Seven-Ingredient Powder for Warming Uterus,the low-dose group of Seven-Ingredient Capsules for Warming Uterus,the high-dose group of Seven-Ingredient Capsules for Warming Uterus.The effects of Seven-Ingredient Formulas for Warming Uterus on the contractile function of rabbit intestinal smooth muscle were analyzed by BL-420 F biological function experimental system.Results:The isolated rabbit intestine experiment showed that low-dose Seven-Ingredient Powder for Warming Uterus and Seven-Ingredient Capsules for Warming Uterus could affect the contractil
