摘 要:为了探究铁、锰离子影响曝气生物滤池(BAF)处理效果的微生物机制,采用最大可能数(MPN)法和高通量测序技术,对比分析了不投加金属离子的BAF-1、投加4.0 mg/L Mn^(2+)的BAF-2,以及同时投加5.0 mg/L Fe^(3+)和4.0 mg/L Mn^(2+)的BAF-3中硝化细菌数量和细菌群落多样性的变化。结果表明:15℃时,BAF-2对COD、NH_(4)^(+)-N、TN和TP的去除率分别比BAF-1提高4、2.14、7.11和10.71个百分点,BAF-3对COD、NH_(4)^(+)-N、TN和TP的去除率分别比BAF-1提高6.93、4.53、8.63和57.29个百分点;同时投加Fe^(3+)和Mn^(2+)比单独投加Mn^(2+)更有利于亚硝酸菌的增殖,Fe^(3+)使受Mn^(2+)影响而减少的硝酸菌反而增多;BAF-2和BAF-3中伯克氏菌目(Burkholderiales)、黄单胞菌目(Xanthomonadales)和根瘤菌目(Rhizobiales)的丰度均高于BAF-1,BAF-3中硝化螺旋菌目(Nitrospirales)、红环菌目(Rhodocyclales)和硫发菌目(Thiotrichales)的丰度均高于BAF-2和BAF-1。研究表明,投加铁、锰离子会使BAF中优势菌群的丰度得到一定程度的增加,有利于强化BAF的处理效果。This study aims at investigating the microbial mechanism of treatment effect changes affected by iron and manganese ions in the biological aerated filter(BAF).The most probable number(MPN)method and high-throughput sequencing technology were used to compare and analyze the number of nitrifying bacteria and bacterial community diversity in three BAFs:BAF-1 without metal ions,BAF-2 with 4.0 mg/L Mn^(2+)as well as BAF-3 with 5.0 mg/L Fe^(3+)and 4.0 mg/L Mn^(2+).The experimental results indicated that when the temperature was 15℃,the removal efficiency of COD,NH_(4)^(+)-N,TN and TP in BAF-2 were 4%,2.14%,7.11%and 10.71%higher than those in BAF-1 respectively.And the removal efficiency of COD,NH_(4)^(+)-N,TN and TP in BAF-3 were 6.93%,4.53%,8.63%and 57.29%higher than those in BAF-1 respectively.Compared with adding Mn^(2+)alone,adding Fe^(3+)and Mn^(2+)together could be more beneficial to the proliferation of nitrosobacteria.The number of nitrobacteria could be decreased by adding Mn^(2+).The number of nitrobacteria could be increased by adding Fe^(3+)and Mn^(2+)together since Fe^(3+)could be beneficial to the proliferation of nitrobacteria.Adding Fe^(3+)and Mn^(2+)together as well as adding Mn^(2+)alone could lead to the increase of bacterial species in biofilm.Adding Mn^(2+)alone could make the abundance of each species more uniform.The abundance of the dominant bacterial community would be higher by adding Fe^(3+)and Mn^(2+)together.The abundance of Burkholderiales,Xanthomonadales,and Rhizobiales in BAF-2 and BAF-3 was higher than that of BAF-1.The abundance of Nitrospirales,Rhodocyclales,and Thiotrichales in BAF-3 was higher than that of BAF-2 and BAF-1.The treatment effect of BAF can be enhanced because the addition of iron and manganese ions can increase the abundance of dominant bacteria in BAF.
