摘 要:目的:观察黄芪多糖对放射性皮炎小鼠模型作用机制。方法:建立放射性皮炎小鼠模型,将80只小鼠随机分为正常组、模型组、黄芪多糖组和三乙醇胺乳膏组,每组20只,连续治疗3周,观察创面愈合情况,苏木精-伊红(HE)染色观察皮肤病理学变化,酶联免疫吸附法(ELISA)检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平,实时荧光定量PCR和蛋白印迹法(Western blot)检测碱性成纤维细胞生长因子(bFGF)、血管内皮生长因子(VEGF)、表皮生长因子(EGF)表达的影响。结果:治疗3周后,黄芪多糖组、三乙醇胺乳膏组小鼠创面愈合率高于模型组,差异均有统计学意义;模型组HE染色见表皮角化,部分表皮内水泡形成,真皮充血水肿,大量炎症细胞浸润;黄芪多糖组、三乙醇胺乳膏组损伤程度明显改善,创面组织修复程度优于模型组;黄芪多糖组和三乙醇胺乳膏组小鼠血清TNF-α、IL-6水平显著降低(P<0.05),皮肤组织中bFGF、VEGF、EGF mRNA水平和蛋白水平均降低(P<0.05),差异均有统计学意义。结论:黄芪多糖能改善皮肤组织中炎症细胞浸润,降低小鼠血清中TNF-α、IL-6水平和皮肤组织中bFGF、VEGF、EGF mRNA水平和蛋白水平,提高创面愈合率。Objective: To observe the action mechanism of Astragalus polysaccharide on radiation dermatitis in mice. Methods: 80 mice were randomly divided into the normal group, the model group, the Astragalus polysaccharide group and the Triethanolamine cream group, with 20 mice in each group. Mice model of radiation dermatitis was established. After 3 weeks of continuous treatment, wound healing was observed, HE staining was used to observe skin pathological changes, ELISA was used to detect the serum levels of tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6), and real-time fluorescent quantitative PCR and Western blot were applied to detect the expressions of basic fibroblast growth factor(bFGF), vascular endothelial growth factor(VEGF) and epidermal growth factor(EGF). Results: After 3 weeks of treatment, the wound healing rate was significantly higher in the Astragalus polysaccharide group and the Triethanolamine cream group than that in the model group. HE staining showed epidermal keratinization, partial intra-epidermal blisters formation, dermal congestion and edema, and a large number of inflammatory cell infiltration in the model group;the Astragalus polysaccharide group and the Triethanolamine cream group showed significant improvement in the degree of injury, and the degree of wound tissue repair was better than that of the model group. The serum levels of TNF-α and IL-6 decreased significantly, and the mRNA and protein levels of bFGF, VEGF and EGF in the skin tissues decreased significantly in the Astragalus polysaccharide group and the Triethanolamine cream group. Conclusion: Astragalus polysaccharides can improve the infiltration of inflammatory cells in skin tissues, reduce serum levels of TNF-α and IL-6, and decrease mRNA and protein expressions of bFGF, VEGF and EGF in skin tissues, and improve wound healing rates.
