摘 要:目的探讨电针对糖尿病胃轻瘫大鼠胃窦Cajal间质细胞(ICC)凋亡的影响。方法取32只SD大鼠,分为空白对照组、模型组、电针组、胃复安组4组,每组8只。采用血糖仪测量大鼠血糖;酚红灌胃法测量计算胃排空率、小肠推进率;提取胃窦ICC原代细胞培养,用Western blot方法检测B细胞淋巴瘤蛋白质2(Bcl-2)、B淋巴细胞瘤-2相互作用蛋白(Beclin1)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的蛋白表达,流式细胞仪检测细胞凋亡率。4组间各指标的比较采用单因素方差分析,组间两两比较采用LSD法。结果与空白对照组相比,模型组、电针组和胃复安组血糖水平均上升,胃排空率、小肠推进率均降低,胃窦ICC Bcl-2、Beclin1、Caspase-3的蛋白表达及细胞凋亡率均升高,差异均有统计学意义(P<0.05)。与模型组相比,电针组、胃复安组血糖水平均下降,胃排空率、小肠推进率均升高,胃窦ICC Caspase-3的蛋白表达及细胞凋亡率均下降,差异均有统计学意义(P<0.05)。与模型组相比,电针组胃窦ICC Bcl-2、Beclin1的蛋白表达均升高(P<0.01)。与胃复安组相比,电针组胃窦ICC Bcl-2、Beclin1的蛋白表达均升高,Caspase-3的蛋白表达及细胞凋亡率均下降(P<0.05)。结论电针可能通过调节凋亡相关因子Bcl-2、Beclin1、Caspase-3的表达而抑制胃窦ICC的凋亡。Objective To explore the effects of electroacupuncture on apoptosis of interstitial cells of Cajal(ICC)in rats with diabetic gastroparesis.Methods Thirty-two SD rats were divided into normal control(NC)group,model group,electroacupuncture group and metoclopramide group,with 8 in each group.The blood glucose was measured by a blood glucose meter.The gastric emptying rate and small intestine propulsion rate were measured by intragastric phenol red method.The primary culture of antral ICC was extracted,the protein expression of B-cell lymphoma protein 2(Bcl-2),B-cell lymphoma-2-interacting protein(Beclin1),Caspase-3 were detected by Western blot,and the apoptosis rate was observed by flow cytometry.One-way analysis of variance(ANOVA)was used for the index comparisons among the 4 groups,and LSD was used for index comparisons between every 2 groups.Results Compared with NC group,the blood glucose was statistically increased,the gastric emptying rate and small intestine propulsion rate were statistically decreased,the protein expression of Bcl-2,Beclin1 and Caspase-3 of ICC and the apoptosis rate of ICC were statistically increased(all P<0.05)in model group,electroacupuncture group and metoclopramide group.Compared with model group,the blood glucose in electroacupuncture and metoclopramide group were statistically decreased,the gastric emptying rate and small intestine propulsion rate were statistically increased,the protein expression of Caspase-3 of ICC and the apoptosis rate of ICC were statistically decreased(all P<0.05).The protein expression of Bcl-2,Beclin1 of ICC in electroacupuncture group statistically increased compared with model group(P<0.01).Compared with metoclopramide group,the protein expression of Bcl-2 and Beclin1 of ICC were statistically increased,and the protein expression of Caspase-3 and the apoptosis rate of ICC in electroacupuncture group were statistically decreased(all P<0.05).Conclusion Electroacupuncture may inhibit the apoptosis of the gastric antrum ICC by regulating the expression of th
