摘 要:目的分析2019年河南省1E基因型风疹病毒分离株结构蛋白基因特征,探讨抗原位点变异情况,为河南省风疹防控提供支持。方法对河南省2019年风疹病毒分离株采用RT-PCR方法扩增结构蛋白基因片段并测序,截取C、E2、E1蛋白序列,使用Mega7.0.26软件进行进化树分析、同源性分析和氨基酸特征分析。结果对5株1E基因型风疹病毒开展研究,扩增结构蛋白基因片段全长3189 bp,同源性分析显示,分离株C、E2、E1蛋白核苷酸序列平均遗传距离分别为0.004、0.002和0.002;与疫苗株(Rubella virus strain BRD-II)序列比较发现,发生氨基酸变异数分别为9个、15个和4个,与抗原相关的位点中,C蛋白103和116位点发生变异,E2蛋白173位点发生变异,E1蛋白333和338位点发生变异,其余关键位点未变异。结论2019年河南省1E基因型风疹病毒株结构蛋白基因中糖基化位点保持稳定,但与细胞免疫相关氨基酸位点发生变异。Objective To analysis the characteristics of structural protein coding gene from 1E genotype rubella virus isolated in Henan province,and explore the variation of the antigenic site for providing the support for rubella control in Henan.Methods The structural protein gene fragment of rubella virus isolated in Henan in 2019 was amplified by RT-PCR,sequenced,and obtained the sequence of C,E2 and E1 protein.The phylogenetic tree analysis,homology analysis,and amino acid characteristic analysis were conducted using MEGA 7.0.26 software.Results Five strains of 1E-genotype virus were selected for this study.The fragment length of structural protein gene amplified was 3189 bp.Homology analysis showed that the average genetic distance of nucleotide sequence of C,E2 and E1 protein was 0.004,0.002 and 0.002,respectively.Compared with the sequence of the vaccine strain(rubella virus strain BRD-II),the number of amino acid variants in the C,E2 and E1 was 9,15 and 4,respectively.Among the antigenic sites,the 103 and 116 sites in C protein,the 173 site in E2 protein,and the 333 and 338 sites in E1protein were mutated,and the other key sites were not mutated.Conclusions The glycosylation sites in structural protein gene of1E-genotype rubella virus strain in Henan were stable,but some mutation took place in the amino acid sites related to cell-mediated immunity.
