摘  要:目的探究动物体内培养软骨微组织-仿生基质复合体的生物学转归。方法采取中华小白猪肋软骨,切割并筛选径长小于200μm的微组织,复合胶原蛋白/透明质酸/硫酸软骨素仿生基质形成软骨微组织-仿生基质复合体,于实验猪体内分别培养2个月、4个月后取出复合体,测量体积变化及吸收率,两组体积采用配对t检验比较。大体标本观察、苏木精-依红染色切片观察其生物学特性及转归。结果体内培养中复合体内软骨微组织可较好存活,纤维组织包裹形成结构稳定的整体,仿生基质可被吸收,移植2个月后平均体积为(2.100±0.115)ml;吸收率为23.333%~36.667%;移植4个月后平均体积为(1.756±0.083)ml;吸收率为36.667%~46.667%,差异有统计学意义(t=10.193,P<0.05)。体内培养过程中出现两种形式细胞增殖,但增殖范围较小,细胞分泌较少。结论动物体内培养软骨微组织-仿生基质复合体可见其具有良好的生物相容性,培养4个月可见少量细胞增殖,但尚无法满足整形外科要求。Objective To preliminarily clarify the biological outcomes of cartilage micro-tissue and biomimetic matrix complex cultured in vivo.Methods The cartilages harvested from experimental Chinese mini-porcine were diced into micro-tissues with diameter less than 200μm.The complexes of cartilage micro-tissue and biomimetic matrix(typeⅡcollagen/hyaluronate acid/chondroitin sulfate)were fabricated and autologous subcutaneous implantation was undergone.After in-vivo culture for 2 months or 4 months,the complexes were harvested.Volume change and absorptivity were measured in terms of(mean±standard deviation),and paired T test was used to compare the volumes of the two groups.All of the harvested complexes underwent Hematoxylin-Eosin staining.The histological characteristics of the complexes and the proliferation and migration of chondrocyte were observed under the microscope.Results The complex had good biocompatibility in vivo.The cartilage microtissue survived with the fibrous tissue proliferating surrounding and encapsulating them to form a stable structure.The biomimetic matrix could be absorbed.The average volume of the complexes at 2nd month after transplantation was(2.100±0.115)ml with the absorption rates ranging from 23.333%to 36.667%,while its counterpart in the other group was(1.756±0.083)ml with the absorption rates being 36.667%to 46.667%.Paired T test was performed,and t=10.193,P<0.05,showing significant difference in the volume between the two groups.Two types of chondrocyte proliferation with small amounts were observed.But the cell proliferations and secretions were limited,which could not meet the requirements of plastic surgery.Conclusion Fine histocompatibility of the complex of cartilage micro-tissue and biomimetic matrix was observed in the in vivoculture.A small amount of cell proliferation was observed after 4 months of culture,but it could not meet the requirements of plastic surgery.
