摘  要:目的观察益气活血复方干预肝癌耐药细胞株HepG2/ADM多药耐药作用,研究其对微环境中P-pg蛋白的影响。方法MTT法检测益气活血复方对HepG2/ADM细胞增长抑制情况及对加入ADM、OXA、5-Fu、索拉菲尼后药敏性的影响,Western blotting法研究其对P-pg蛋白的影响。结果经益气活血方处理24~72 h,HepG2及HepG2/ADM抑瘤率均升高,与时间及浓度正相关,二者差异无统计学意义(P>0.05),分别加入ADM、OXA、5-Fu、索拉菲尼,抑制增殖效果仍明显,中、高剂量组较对照组差异有统计学意义(P<0.05)。随药物浓度增加,P-gp蛋白表达明显降低(P<0.05)。结论益气活血复方可抑制HepG2/ADM的增殖,且能抑制ADM、OXA、5-FU、索拉菲尼的耐药性,其机制可能与降低肿瘤微环境中耐药相关基因P-gp蛋白有关。Objective To study the effect of benefiting qi for activating blood circulation compound on the proliferation of HepG2/ADM and its effect on P-pg proteins in the microenvironment.Methods MTT method was used to detect the inhibition rate of different benefiting qi for activating blood circulation compound concentrations on HepG2/ADM resistant cells and its effect on drug sensitivity of resistant cells with ADM/OXA/5-Fu/Sorafenib.Western blotting method was used to detect the expression of P-pg proteins.Results Liver cancer HepG2 and liver cancer HepG2/ADM resistant cells were significantly lower than before after 24h to 72h with benefiting qi for activating blood circulation compound.Inhibition rate was positively correlated with time and concentration.There was no significant difference in inhibition between the two groups(P>0.05).The inhibition effect of cell proliferation was still obvious with ADM/OXA/5-Fu/Sorafenib.The middle and high dose groups were significantly lower than the control group(P<0.05).The P-gp protein was gradually reduced in the microenvironment with the concentration of benefiting qi for activating blood circulation compound gradually increased,and the difference was statistically significant(P<0.05).Conclusion Benefiting qi for activating blood circulation compound can significantly inhibit the proliferation of HepG2/ADM resistant cells in vitro.The mechanism is related to the reduction of P-gp protein in tumor microenvironment.
