摘 要:绵羊脱毛症受环境和基因的因素共同决定,在分子育种日益盛行的时代,筛选脱毛症相关分子标记,可为绵羊的健康养殖提供参考数据。采取100只健康中国美利奴羊为对照组和50只脱毛症中国美利奴羊为病例组,提取其DNA,制备基因组芯片,采用PLINK软件质量控制以提取OAR1(Ovis aries 1绵羊1号染色体),通过Vcftools对所获得的数据进行遗传分化系数(F_(st))计算,从大到小选择总数值的前1%。结果表明:经过质量控制后,获得相关SNPs为4958个;F_(st)计算后获得分子标记35个,经过基因注释获得53个,其中影响皮肤发育的基因有GADD45A、MCCC1、MLPH、SSBP3等,已经有文献报道SSBP3与脱毛症相关。对获得的分子标记可以为绵羊分子选育提供参考,缩短世代间隔,提高育种年限,节约成本,最终获得免疫力强的健康绵羊,防止绵羊脱毛症的出现,从而减少农牧民的经济损失。Sheep alopecia is determined by both environmental factors and genetic factors.In the era of increasing prevalence of molecular breeding,screening molecular markers related to depilation can provide reference data for sheep healthy breeding.In this experiment,100 healthy Chinese Merino sheep were used as the control group,and 50 Chinese Merino sheep with depilation disease were used as the case group to extract DNA,prepare genomic chip,extract oar1(Ovis aries 1 chromosome 1)by PLINK software of the quality control,calculate the genetic differentiation coefficient(F_(st))of the obtained data through Vcftools and select the first 1%of the total value from large to small.The results showed that 4958 SNPs were obtained after quality control,and 35 molecular markers were obtained after F_(st) calculation.53 genes were obtained after gene annotation,among which GADD45A,MCCC1,MLPH and SSBP3 were the genes affecting skin development.It has been reported that SSBP3 is related to depilation.The obtained molecular markers can provide reference for sheep molecular breeding,shorten generation interval,improve breeding period,save cost,finally obtain healthy sheep with strong immunity,prevent the occurrence of sheep depilation,and reduce the economic loss of farmers and herdsmen.
