摘 要:为探讨激素调节太子参块根形成的机制,通过对不同发育时期的太子参块根进行比较转录组分析,将块根形成初期、块根迅速膨大期和块根成熟期分别与不定根形成期进行比较,从3个比较组(不定根形成期与块根形成初期、不定根形成期与块根迅速膨大期、不定根形成期与块根成熟期)中分别获得5132、5506、22709个差异表达基因.通过韦恩图进一步筛选,在不定根形成期与块根形成初期组中,根据特异上调或下调的差异表达基因挖掘块根形成过程中的关键调控因子,不定根形成期与块根形成初期组共有1215个特异性差异表达基因,其中858个基因特异性差异上调表达,357个基因特异性差异下调表达.KEGG途径富集分析表明,在不定根形成期与块根形成初期组中,特异性差异表达的激素合成及信号转导途径相关基因共27个,其中玉米素生物合成途径相关基因1个,植物激素信号转导途径相关基因26个,涉及生长素、细胞分裂素、赤霉素、乙烯和茉莉酸等信号通路.To investigate the mechanism of hormone-regulated tuberous root formation in Pseudostellaria heterophylla,comparative transcriptome analysis of the roots was conducted at different developmental stages.In comparison to the adventitious root(ARs),5132,5506 and 22709 differentially expressed genes were obtained from the initiated tuberous roots(ITRs),medium tuberous roots(MTRs)and later tuberous roots(LTRs),respectively.For the exploration of key regulatory factors during tuber formation,a total of 1215 differentially expressed genes(858 up-regulated and 357 down-regulated)were specifically screened in the ARs_VS_ITRs by Venn diagram.KEGG pathway enrichment analysis further revealed that 27 differentially expressed genes were related to hormone synthesis and signal transduction,among which 1 gene participated in zeatin biosynthetic pathway and 26 genes were involved in hormone signal transduction,concerning signaling pathways regulating auxin,cytokinin,gibberellin,ethylene,jasmonic acid,etc.
