摘  要:目的:对金莲花抗肠道病毒71型(enterovirus 71,EV71)物质基础进行研究。方法:通过建立超高效液相色谱-串联二极管阵列检测器-飞行时间质谱(UPLC-DAD-TOF/MS)指纹图谱对金莲花物质基础进行表征,通过噻唑蓝(MTT)法对金莲花抗EV71病毒抑制率进行检测,通过正交偏最小二乘法(OPLS)和灰色关联分析法(GRA)对金莲花物质基础和病毒抑制率进行谱效相关关联分析。结果:建立了20批次金莲花样品的指纹图谱,标定了31个共有峰,并进行了质谱解析;建立了金莲花对EV71抑制率检测方法,20批次金莲花的病毒抑制率为49.64%~73.69%;OPLS和GRA分析结果表明,共有峰12、13、14、18可能为金莲花抗EV71物质基础。结论:共有峰12(番石榴酸)、13(未鉴定的生物碱成分)、14(对羟基苯甲苹果酸)和18(2″-O-乙酰基荭草苷)可能为金莲花抗EV71物质基础,此可为金莲花质量标准的建立和金莲花抗病毒研究提供借鉴和数据支持。Objective:To study the anti-enterovirus 71(EV71) material basis of Jinlianhua(Trollius Chinensis Bge.).Methods:The material basis was characterized by establishing UPLC-DAD-TOF/MS fingerprint of Jinlianhua,the virus inhibition rate of Jinlianhua against EV71 was detected by MTT method,and the spectrum-effect correlation analysis on material basis and virus inhibition rate of Jinlianhua was performed by orthogonal partial least squares(OPLS) and grey relational analysis(GRA).Results:The fingerprints of 20 batches of Jinlianhua samples were established,31 common peaks were marked,and the mass spectrometry analysis was analyzed.The detection method of EV71 inhibition rate of Jinlianhua was established,and the virus inhibition rates of 20 batches of Jinlianhua were 49.64%~73.69%.OPLS and GRA analysis results indicated that the common peaks 12,13,14,and 18 may be the material basis of anti-EV71 in Jinlianhua.Conclusions:The common peaks 12(piscidic acid),13,14(2-(4-hydroxybenzyl) malic acid),and 18(2''-Oacetylorientin) may be the material basis of anti-EV71 in Jinlianhua.This can provide reference and data support for the establishment of quality standard and anti-virus research of Jinlianhua.
