摘 要:foxl2是性腺分化、发育和维持等发育过程的转录调节因子,在一些哺乳动物和鱼类的卵巢发育中发挥了重要作用。本研究比较分析了性成熟前后罗氏沼虾(Macrobrachium rosenbergii)foxl2基因在肝胰腺与卵巢中的表达,并利用RNA干扰技术沉默MrFoxl2(M.rosenbergii foxl2)基因后分析其在卵巢成熟过程中的功能。荧光定量结果显示,MrFoxl2在卵巢组织中高表达,并且性成熟后MrFoxl2在卵巢中的表达较性成熟前显著提高。在注射MrFoxl2-dsRNA后MrFoxl2基因的表达显著下降,并且使用1μg/g的注射剂量对MrFoxl2基因的敲降效果最佳。此外,MrFoxl2基因沉默后,生殖相关基因VG与EcR的表达显著下调,CatL基因表达显著上调。研究结果表明,MrFoxl2在罗氏沼虾卵巢成熟过程中可能发挥了重要作用。The hepatopancreas and ovaries of immature and matured Macrobrachium rosenbergii were collected respectively to investigate the functions of forkhead box L2(foxl2)in the reproductive process in female giant freshwater prawns(M.rosenbergii).The mRNA expression of MrFoxl2(M.rosenbergii foxl2)was analyzed by quantitative real-time PCR(qPCR).The negative control nuclease-free water and three different doses(0.5μg/g,1μg/g,and 2μg/g of body weight)of MrFoxl2-dsRNA were injected into the second abdominal segment of M.rosenbergii to investigate the knockdown performance on MrFoxl2.Changes in expression of reproduction-related genes(VG、EcR,and CatL)were detected after foxl2 interference.The results suggested that the expression level of MrFoxl2 in matured ovaries was significantly higher than that of testis immature ovaries.The expression of MrFoxl2 in ovaries was significantly down-regulated after three different doses MrFoxl2-dsRNA injection and 1μg/g was optimal dose.The MrFoxl2 expression was further decreased as the injected dose increased from 0.5μg/g to 1μg/g,and relative increase in MrFoxl2 expression as the injected dose increased from 1μg/g to 2μg/g.After MrFoxl2 interference,the expression of VG and EcR was significantly down-regulated and CatL expression significantly increased.However,as the injection dose increased from 0.5μg/g to 1μg/g,the expression levels of VG and EcR genes increased relatively,which was different from the changes in MrFoxl2 expression.Therefore,our results suggested that MrFoxl2 plays an important role in the ovarian maturation of M.rosenbergii.The current study provides references for the studies on reproductive regulation of M.rosenbergii.
