摘 要:为研究大口黑鲈(Micropterus salmoides)对葡萄糖的耐受能力和糖代谢关键酶的影响,选取96尾体质量为165.42±5.45 g的大口黑鲈禁食24 h,设置对照组(C组,0.8%生理盐水)和实验组(G组,注射2g/kg体重葡萄糖)。结果显示:注射葡萄糖后,血糖水平先上升后下降,在8 h达到最高水平,直至48 h恢复至基础水平。G组肝糖原含量、己糖激酶(HK)和丙酮酸激酶(PK)活性先上升后下降,且均高于对照组。与对照组相比,G组血液、肝脏和肌肉中乳酸(LD)含量均有一定程度的升高,Na^(+)-K^(+)-ATP酶的活性显著升高。G组肝脏糖异生关键酶磷酸烯醇式丙酮酸羧激酶(PEPCK)活性在1 h显著上升,然后持续下降并且在12 h显著低于C组。G组AMP依赖的蛋白激酶α(AMPKα)和葡萄糖转运体(Glut 1)的基因表达量低于C组;G组Glut 4的表达量在注射后的短时间内上调。以上结果表明,大口黑鲈对葡萄糖的耐受能力较低。注射葡萄糖加强其糖酵解过程并促进了乳酸和糖原生成,而对糖异生途径的调控能力较差。A total of 96 Micropterus salmoides(165.42±5.45)g were selected to design two groups,control(C,0.8%normal saline)and glucose injection(G,2g/kg body weight)after fasted for 24h to study the effects on glucose tolerance and the key enzymes of glucose metabolism in M.salmoides.The results showed that the plasma glucose level rose firstly and then decreased,reaching the highest level at 8h,and returning to the base level at 48h in group G.Similarly,the liver glycogen content,hexokinase(HK)and pyruvate kinase(PK)activities of group G increased at first and then decreased,but they were higher than those indexes in group C.Compared with group C,the content of lactic acid(LD)presented increase tendency significantly in plasma,liver and muscle of group G,including the activity of Na^(+)-K^(+)-ATPase.Hepatic gluconeogenesis key enzyme of phosphoenolpyruvate carboxykinase kinase(of PEPCK)activity was significantly increased at 1 h in group G,then continued to decline and was significantly lower than that of group C at 12 h.The gene expression of AMP-dependent protein kinaseα(AMPKα)and glucose transporter(Glut1)were regulated in group G.However,GLUT4 expression was negative up-regulated in a short time after injection.These results indicated that M.salmoides has a low glucose tolerance.Glucose injection could strengthen the glycolysis process and promote the production of lactic acid and glycogen production in M.salmoides,but it expresses the low ability to regulate gluconeogenesis.
