摘 要:目的探讨滨蒿总黄酮(Total flavonoids from Artemisia scoparia Waldst.et Kit.,ASTF)对抗流感病毒作用及H1N1流感病毒感染的人肺癌A549细胞先天免疫反应过程中起调控作用。方法定量检测H1N1流感病毒致犬肾上皮MDCK细胞病变(Cytopathic effect,CPE)作用;用TRIzol提取总RNA,RT-qPCR检测ASTF对流感病毒相关信号通路的抗病毒效果,并用流式微球技术(Cytometric bead array,CBA)和ELISA检测ASTF对H1N1流感病毒感染的A549细胞分泌炎性细胞因子的影响。结果ASTF在体外对H1N1流感病毒有一定抑制作用,可降低H1N1感染的A549细胞先天免疫通路中的RIG-I、AKT、P38、PI3K、STING、PKR和Viperin mRNA表达水平及炎性细胞因子IL-2、IL-6、IL-17A的分泌。结论ASTF通过抑制胞内先天免疫信号通路RLRs、PI3K/AKT、P38 MAPK发挥免疫调节和抗炎作用,从而达到抗流感的效果。Objective To investigate the anti-influenza effects and regulatory mechanism of total flavonoids from Artemisia scoparia Waldst.et kit.(ASTF)on the innate immune signaling pathway of human lung adenocarcinoma cell line A549 infected by H1N1 influenza virus.Methods Viral ToxGlo was used to evaluate the cytopathic effect(CPE)of H1N1 influenza virus on the canine renal epithelial cells MDCK and the anti-influenza H1N1 virus effect of ASTF;Total RNA was extracted by TRIzol method,and then reverse transcription fluorescence quantitative PCR(RT-qPCR)was performed to detect the regulatory mechanism of ASTF on innate immune signaling pathway in human lung adenocarcinoma cell line A549 infected by H1N1 influenza virus.The cytometric bead array(CBA)and ELISA method were further exploited to detect the impacts of ASTF on the secretion of inflammatory cytokines in A549 cells infected by H1N1 influenza virus.Results ASTF showed some inhibitory effects against H1N1 influenza virus in vitro.It could decrease the mRNA expression levels of RIG-I,AKT,P38,PI3K,STING,PKR and Viperin as well as the inflammatory secretion of IL-2,IL-6 and IL-17A in A549 cells infected by H1N1 influenza virus.Conclusion ASTF plays an immunomodulatory and anti-inflammatory role via inhibiting the innate immune signaling pathway of RLRs,PI3K/AKT,P38 MAPK,thus achieving the anti-influenza effects.
