摘  要:为探究MYB4b基因在调控植物生长发育以及对外界环境的响应,以‘摩尔多瓦’葡萄为材料,通过PCR克隆得到VvMYB4b基因序列,通过在线软件分析其基本序列特征,采用荧光定量PCR方法检测VvMYB4b基因在‘摩尔多瓦’不同组织部位和不同农艺技术处理下的转录表达水平,同时检测了相应的花色苷含量。结果发现,从‘摩尔多瓦’葡萄中克隆到VvMYB4b转录因子的序列全长729 bp,编码242个氨基酸,属于亲水性的非分泌碱性蛋白,系统进化树显示其与拟南芥的AtMYB4同源性较高。VvMYB4b在葡萄不同组织中均有表达,其中表达量最高的组织为茎尖,表达量最低的组织为幼果期果实;各农艺技术处理果实均显示下调VvMYB4b基因的表达,其中0.3%盐和壳聚糖的下调表达更强烈。由此推测,VvMYB4b可能参与调控‘摩尔多瓦’葡萄生理环境应答过程,为进一步研究葡萄VvMYB4b基因的功能奠定了基础。In order to explore the important role of MYB4b gene in regulating plant growth and development and the response process to external environment,this study took'Moldova'as material,cloned the VvMYB4b gene sequence by PCR,and analyzed its basic sequence characteristics by online software.The transcription and expression levels of VvMYB4b gene in different tissue parts of'Moldova'and under different treatments were detected by fluorescence quantitative PCR,and the corresponding anthocyanin content was detected.The results showed that the sequence of VvMYB4b transcription factor was cloned from'Moldova',with a total length of 729 bp,encodes 242 amino acids,and belongs to hydrophilic non secretory basic protein;Phylogenetic tree showed that VvMYB4b gene had high homology with AtMYB4 of arabidopsis thaliana;VvMYB4b was expressed in different tissues of grape,among which the highest expression tissue was the stem tip,and the lowest expression tissue was the fruit at the young fruit stage;The expression of VvMYB4b gene was down regulated by 0.3%salt,PEG6000,Bihu and chitosan,and the down regulated expression was stronger by 0.3%salt and chitosan.It is speculated that VvMYB4b may play a role in the regulatory response of'Moldova'during physiological stress,which lays a foundation for further study on the function of'Moldova'VvMYB4b gene.
