摘  要:目的观察天麻芎苓止眩片对自发性高血压大鼠血清炎症因子及胸主动脉组织Atg8、LC3及p62表达的影响,探讨其治疗高血压的作用机制。方法将30只自发性高血压大鼠随机分为模型组、卡托普利组和天麻芎苓止眩片组,每组10只,另设10只WKY大鼠为正常对照组。卡托普利组和天麻芎苓止眩片组分别以7.6、43.2 mg/kg相应药物灌胃,正常对照组及模型组予蒸馏水灌胃,给药体积均为10 mL/kg,每日1次,连续8周。测量大鼠血压,ELISA检测大鼠血清白细胞介素(IL)-1、IL-10含量,HE染色观察大鼠胸主动脉组织病理变化,免疫组化、Western blot和RT-PCR检测大鼠胸主动脉组织Atg8、LC3、p62蛋白和mRNA表达。结果与正常对照组比较,模型组大鼠血压明显升高(P<0.05),血清IL-1含量明显增加,IL-10含量明显减少(P<0.05);胸主动脉管壁增厚,内膜损伤明显;胸主动脉组织Atg8、LC3、p62蛋白表达及LC3Ⅱ/LC3Ⅰ比值明显升高(P<0.05),Atg8、LC3Ⅱ、p62 mRNA表达明显升高(P<0.05)。与模型组比较,给药第3周开始,天麻芎苓止眩片组大鼠血压明显降低(P<0.05),血清IL-1含量明显减少(P<0.05),IL-10含量明显增加(P<0.05);胸主动脉管壁增厚有所改善,内膜损伤减轻;胸主动脉组织Atg8、LC3、p62蛋白表达及LC3Ⅱ/LC3Ⅰ比值明显降低(P<0.05),Atg8、LC3Ⅱ、p62 mRNA表达明显降低(P<0.05)。结论天麻芎苓止眩片可能通过减轻炎症反应、抑制内皮细胞过度自噬、保护内皮功能,发挥治疗高血压的作用。Objective To observe the effects of Tianma Xiongling Zhixuan Tablets on the serum inflammatory factors,and expressions of Atg8,LC3 and p62 in thoracic aorta tissue with spontaneously hypertensive rats;To explore the potential mechanism of treating hypertension.Methods Totally 30 spontaneously hypertensive rats were randomly divided into model group,captopril group and Tianma Xiongling Zhixuan Tablets group,with 10 rats in each group,and 10 WKY rats as normal control group.The captopril group and the Tianma Xiongling Zhixuan Tablets group were given 7.6 and 43.2 mg/kg corresponding medicine by gavage respectively.The normal control group and the model group were given distilled water at a volume of 10 mL/kg for gavage,once a day,for 8 consecutive weeks.The blood pressure was measured,ELISA was used to detect the contents of interleukin(IL)-1 and IL-10 in serum.HE staining was used to observe the pathological changes of thoracic aorta tissue.Immunohistochemistry,Western blot and RT-PCR were used to detect Atg8,LC3 and p62 protein and mRNA expression in thoracic aorta tissue.Results Compared with the normal control group,the blood pressure of rats in the model group significantly increased(P<0.05),the content of IL-1 in serum significantly increased(P<0.05),the content of IL-10 significantly reduced(P<0.05).The wall of the thoracic aorta was thickened and the intimal injury was obvious.The expressions of Atg8,LC3,p62 protein and the ratio of LC3Ⅱ/LC3Ⅰin thoracic aorta tissue significantly increased(P<0.05),the expressions of Atg8,LC3Ⅱ,p62 mRNA significantly increased(P<0.05).Compared with the model group,the blood pressure of the rats in Tianma Xiongling Zhixuan Tablets group significantly decreased after three weeks of administration(P<0.05),the content of IL-1 in serum significantly reduced(P<0.05),the content of IL-10 significantly increased(P<0.05).The thickening of thoracic aortic wall was improved,the intimal damage was reduced.The expressions of Atg8,LC3,p62 protein and the ratio of LC3Ⅱ/LC3Ⅰin thor
