摘 要:目的观察益气温阳方对变应性鼻炎(AR)小鼠鼻黏膜线粒体动态变化及活性氧(ROS)的影响,探讨其干预AR的潜在机制。方法卵清蛋白腹腔注射联合滴鼻强化致敏制备AR小鼠模型,将小鼠随机分为空白组、模型组、益气温阳方组及氯雷他定组,每组10只,分别予相应药液灌胃。评估小鼠行为学变化,HE染色观察鼻黏膜组织形态,ELISA测定血清白细胞介素(IL)-4、IL-5、γ干扰素(IFN-γ)含量;荧光探针法检测鼻黏膜组织ROS水平,电镜观察鼻黏膜线粒体形态。结果与空白组比较,模型组小鼠搔鼻、喷嚏次数明显增多(P<0.01),鼻黏膜上皮破坏明显,纤毛紊乱,黏膜下可见大量嗜酸性粒细胞浸润,间质存在增生及水肿;血清IL-4、IL-5含量明显增加,IFN-γ含量明显减少(P<0.01),鼻黏膜组织ROS水平明显升高(P<0.01);线粒体呈圆球状分布,线粒体脊、双层膜结构较模糊。与模型组比较,益气温阳方组小鼠搔鼻、喷嚏次数明显减少(P<0.01),鼻黏膜上皮损伤、纤毛紊乱程度、间质增生及水肿有所改善,黏膜下嗜酸性粒细胞数量明显减少;血清IL-4、IL-5含量明显减少(P<0.05,P<0.01),IFN-γ含量明显增加(P<0.01),鼻黏膜组织ROS水平明显降低(P<0.01);线粒体形态有所恢复,线粒体长度增加,上述效应均与氯雷他定组相当。结论益气温阳方干预AR的潜在效应可能与调节鼻黏膜线粒体动态变化及ROS内生相关。Objective To observe the effects of Yiqi Wenyang Prescription on the dynamic changes of mitochondria and ROS in nasal mucosa of allergic rhinitis(AR)mice;To explore the potential mechanism of its intervention in AR.Methods Allergic rhinitis mice model were established by intraperitoneal injection of ovalbumin combined with nasal drops to enhance sensitization,the mice were randomly divided into blank group,model group,Yiqi Wenyang Prescription group and Loratadine group,with 10 mice in each group.Each group was given corresponding medicine for gavage.Behavioral changes of mice were evaluated;the pathological changes of nasal mucosa tissue were observed by HE staining;the contents of serum IL-4,IL-5,and IFN-γwere detected by ELISA;the level of ROS in nasal mucosa tissue was detected by fluorescent probe;morphological changes of nasal mucosa mitochondria was observed by electron microscope.Results Compared with the blank group,the number of sniffing and sneezing of mice in the model group increased significantly(P<0.01).The epithelium of the nasal mucosa was damaged obviously.The cilia were disordered,a large number of eosinophils were infiltrated under the mucosa,there was hyperplasia and edema in the interstitium.The contents of IL-4 and IL-5 in serum significantly increased,the content of IFN-γsignificantly decreased(P<0.01).The level of endogenous ROS in nasal mucosa tissue increased(P<0.01).The mitochondria were distributed in a spherical shape,the mitochondrial ridges and bilayer membrane structures were blurred.Compared with the model group,the number of sniffing and sneezing of mice in Yiqi Wenyang Prescription group significantly reduced(P<0.01).The damage of nasal mucosal epithelial,the degree of cilia disorder.The hyperplasia and edema of interstitial were significantly improved,the number of submucosal eosinophils reduced.The contents of IL-4 and IL-5 in serum decreased(P<0.05,P<0.01),the content of INF-γincreased(P<0.01).The level of endogenous ROS in nasal mucosal tissue decreased(P<0.01).The morph
