摘 要:以云南民间药用植物青阳参为研究材料,利用超声提取法处理,HPLC法检测,考察青阳参苷元提取率.基于单因素实验结果,对乙醇体积分数、超声温度、超声时间采用Box-Behnken实验设计和响应面法(RSM)优化提取条件,从而获得最佳提取工艺参数,即提取溶液的盐酸浓度0.36 mol/L、料液比1∶10(g/mL)、乙醇体积分数58%、超声功率600 W、超声温度63℃、超声时间33 min,验证提取得到产物青阳参苷元提取率0.2745%,实验结果与模型预测值接近,表明最佳工艺参数可行.The Yunnan folk medicinal plant Cynanchum otophyllum Schnei were used as materials,and the extraction rate of Qingyangshengenin were investigated by the ultrasonic extraction process and HPLC.Based on the single factor experimental results of ethanol concentration,ultrasonic temperature and ultrasonic time,Box Behnken experimental design and response surface methodology(RSM)were selected to further analyze and optimize the extraction conditions.The finnal extraction process was hydrochloric acid concentration 0.36 mol/L,solid-liquid ratio 1∶10(g/mL),58%ethanol,600 W ultrasonic power,63℃ultrasonic temperature and 33 min ultrasonic time.The optimum extraction rate of Qingyangshengenin was 0.2745%.The experimental results are close to the predicted values of the model,indicating that the optimal process conditions are feasible.
