摘　　要：目的:探索复方黄柏液对内皮细胞增殖迁移成管影响的机制,为复方黄柏溶液用于治疗糖尿病足溃疡提供依据。方法:用CCK-8法检测药物毒性,探索复方黄柏液使用的最佳药物浓度、CCK-8法检测细胞增殖活力、细胞划痕实验检测细胞迁移率、成管实验检测细胞成管能力、蛋白质印迹法(Western Blotting)实验检测蛋白表达水平的变化。结果:使用复方黄柏液后,高糖组内皮细胞的增殖、迁移、成管能力较前有了提高,并降低了促血管生成素2(Ang-2)蛋白水平,增加血管内皮生长因子(VEGF)和磷酸化酪氨酸激酶受体-2(P-Tie)蛋白水平。结论:复方黄柏液通过促进内皮细胞增殖、迁移、成管以及影响蛋白表达水平,改善内皮功能障碍来实现对糖尿病足溃疡的恢复。Objective:To explore the effect and mechanism of Fufang Huangbai Liquid on endothelial cell proliferation,migration,and tube formation and provide evidence for the treatment of diabetic foot ulcer by Fufang Huangbai Liquid.Methods:Cell Counting Kit-8(CCK-8)was used for the detection of drug toxicity to determine the optimal concentration of Fufang Huangbai Liquid.Cell proliferation activity was detected by CCK-8 assay,cell migration rate by wound healing assay,tube formation ability by tube formation assay,and protein expression changes by Western blot.Results:After Fufang Huangbai Liquid treatment,the proliferation,migration,and tube formation abilities of endothelial cells in the high-glucose group were potentiated.Furthermore,the angiopoietin 2(Ang2)protein expression was reduced,and the protein expression levels of vascular endothelial growth factor(VEGF)and phosphorylated tyrosine kinase receptor-2(p-Tie2)were up-regulated.Conclusion:Fufang Huangbai Liquid can relieve diabetic foot ulcer by promoting endothelial cell proliferation,migration,and tube formation,affecting protein expression,and improving endothelial dysfunction.
