摘  要:类黄酮-3′,5′-羟基化酶(Flavonoid 3′,5′-hydroxylase,F3′5′H)是类黄酮代谢途径的一个关键酶,能使花色素的合成方向趋向于蓝色的飞燕草素。利用生物信息学软件对锦绣杜鹃类黄酮-3′,5′-羟基化酶基因(RpF3′5′H)进行分析。结果显示,RpF3′5′H基因ORF长1551 bp,共编码516个氨基酸;分子量为57.31 kD,等电点为9.03,无信号肽,定位于细胞质中的可能性最大,二级结构与三级结构中含有较多的α螺旋,与高丛越橘有较高亲缘性。分析结果将对RpF3′5′H的功能解析奠定理论基础,同时也为探究锦绣杜鹃花色苷的生物合成有一定指导作用。Flavonoid 3′,5′-hydroxylase(F3′5′H)is a key enzyme in flavonoid metabolism pathway,which can make the biosynthesis of anthocyanins tend to blue delphinidin.F3′5′H gene of Rhododendron pulchrum was analyzed by bioinformatics software.The results showed that ORF of RpF3′5′H gene was 1551 bp,encoding 516 amino acids,with a molecular weight of 57.31 KD and an isoelectric point of 9.03.Signal peptide was not detected in RpF3′5′H and it was likely located in the cytoplasm.There were majorα-helix in the secondary and tertiary structures of RpF3′5′H,meanwhile,RpF3′5′H had high affinity with blueberry.The results will lay a theoretical foundation for the functional analysis of RpF3′5′H,and also have a certain guiding role in the study of anthocyanin biosynthesis of Rhododendron pulchrum.
