不同浓度丙泊酚调控H_(2)O_(2)损伤心肌细胞的相关通路探究

摘  要:目的分析不同浓度丙泊酚调控H_(2)O_(2)损伤心肌细胞的相关通路。方法于5%CO_(2)、37℃下使用含10%胎牛血清的DMEM培养基培养H9c2细胞,选取对数生长期细胞开展后续实验,共设置5组对照组实验。其中对照组细胞不给予任何处理,模型组细胞给予200μmol/L终浓度的H_(2)O_(2)培养24 h,A-低浓度组、中浓度组、高浓度组分别给予12.5、25.0、50.0μmol/L丙泊酚,同时给予200μmol/L终浓度的H_(2)O_(2)培养24 h。对各组细胞培养24 h后细胞存活、凋亡、侵袭、迁移情况进行比较,并对各组细胞培养24 h后抗氧化指标[乳酸脱氢酶(LDH)、丙二醛、活性氧、过氧化氢酶(CAT)、谷胱甘肽(GSH)]水平、炎症因子[白细胞介素-6(IL-6)、一氧化氮(NO)、肿瘤坏死因子α(TNF-α)]水平、血红素氧合酶-1(HO-1)及核因子E2相关因子2(Nrf2)蛋白及mRNA水平进行比较。结果与对照组相比,模型组细胞存活率均降低,凋亡率均升高,差异均有统计学意义(P<0.05);与模型组相比,低浓度组、中浓度组、高浓度组细胞存活率均升高,凋亡率均降低,差异均有统计学意义(P<0.05)。与对照组和模型组相比,低浓度组、中浓度组、高浓度组细胞侵袭个数和迁移个数均减少,差异均有统计学意义(P<0.05)。与对照组相比,模型组细胞LDH、丙二醛、活性氧、IL-6、TNF-α水平均升高,CAT、GSH、NO水平均降低,差异均有统计学意义(P<0.05);与模型组相比,低浓度组、中浓度组、高浓度组细胞LDH、丙二醛、活性氧、IL-6、TNF-α水平均降低,CAT、GSH、NO水平均升高,差异均有统计学意义(P<0.05)。与对照组相比,模型组细胞HO-1、Nrf2及其mRNA水平均降低,差异均有统计学意义(P<0.05);与模型组相比,低浓度组、中浓度组、高浓度组细胞HO-1、Nrf2及其mRNA水平均升高,差异均有统计学意义(P<0.05),且以上作用呈浓度依赖性。结论丙泊酚能够通过调控Nrf2/HO-1通路抑制H_(2)O_(2)诱导心肌细�Objective To analyze the relevant pathways of different concentrations of propofol regulating H_(2)O_(2)damage to cardiomyocytes.Methods H9c2 cells were cultured in DMEM medium containing 10%fetal bovine serum under 5%CO_(2),37℃,and logarithmic growth phase cells were selected for follow-up experiments.A total of 5 groups of control groups were set up.The control group cells were not given any treatment,and the model group cells were cultured with 200μmol/L final concentration of H_(2)O_(2)for 24 h.Low concentration group,medium concentration group,and high concentration group were given 12.5μmol/L and 25.0μmol/L,50.0μmol/L,respectively,while giving 200μmol/L final concentration of H_(2)O_(2)for 24 h.The cell survival,apoptosis,invasion and migration of each group of cells after 24 hours of culture were compared,and the levels of antioxidant indexes[lactate dehydrogenase(LDH),malondialdehyde(MDA),reactive oxygen species,catalase(CAT),glutathione(GSH)],inflammatory factors[interleukin-6(IL-6),nitric oxide(NO),tumor necrosis factor-α(TNF-α)],and heme oxygenase-1(HO-1)and nuclear factor E2 related factor 2(Nrf2)protein and mRNA levels of each group of cells after 24 hours of culture were compared.Results Compared with the control group,the cell survival rate of the model group decreased,and the apoptosis rate increased,and the differences were statistically significant(P<0.05);compared with the model group,the survival rate of the cells in the low concentration group,the medium concentration group and the high concentration group increased,and the apoptosis rate decreased,and the differences were statistically significant(P<0.05).Compared with the control group and the model group,the number of invasion and migration of cells in the low concentration group,the medium concentration group and the high concentration group decreased,and the differences were statistically significant(P<0.05).Compared with the control group,the levels of LDH,malondialdehyde,reactive oxygen species,IL-6,and TNF-αin the model grou
