摘  要:目的反向微生态学(RM)方法预测与类风湿性关节炎(RA)发病相关的肠道微生态蛋白抗原,证明RM方法在微生态学研究的可行性。方法OHMI本体标准化表述和分析RA相关的肠道微生态菌种,提取代表菌株的全基因序列,RM方法预测与RA发病相关蛋白抗原。结果本体分析发现25个与RA相关的肠道微生态菌种,其中致病菌14株、益生菌11株。通过对8个菌种的27个全基因分析,预测了可能与RA发病相关的人同源蛋白10个和非同源蛋白330个。结论RM可快速预测疾病相关的肠道微生态病原性自身抗原和毒力因子,可在分子水平探讨微生态的致病机制。Objective To predict rheumatoid arthritis-associated gut microbiota pathogenic antigens using ontology and reverse microbiomics(RM),and to certificate the feasibility of the RM in microbiome study.Methods OHMI was used for standard formulation and analysis RA-associated gut microbiome.The genome sequences of bacterial species were retrieved,and RM strategy was used to predict protein antigens related to RA morbidity.Results Our ontology identified 25 intestinal microecological species that were related to RA morbidity.Out of these 25 bacteria,14 were pathogenic bacteria,and 11 probiotics.We predicted a total of ten bacterial proteins had homology to human proteins and 330 non-human homology proteins that were likely associated with RA through analysis 27 whole genes of 8 strains.Conclusions RM could rapidly predict pathogenic auto-antigens and virulence factors of intestinal microecology associated with disease,and could explore the pathogenic mechanism of microecology at the molecular level.
