摘  要:在庆大霉素诱导的不同程度急性肾损伤大鼠模型静脉注射鸡尾酒转运体标记物,通过对各组大鼠肾脏相关转运蛋白表达水平和血清及尿液中底物浓度变化差异进行综合评价.研究发现,中低剂量组大鼠的肾小球滤过功能代偿性增强,肾小管分泌功能轻度下降;随着损伤程度的加重,大鼠的肾小球滤过及肾小管分泌功能明显降低.75及100 mg/(kg·日)庆大霉素组大鼠血清肌酐、五氟尿嘧啶、呋塞米、二甲双胍及对氨基马尿酸的血药浓度-时间曲线下面积升高,除呋塞米外的各个底物清除率均有不同程度的下降.The rat model for acute kidney injury induced by gentamicin(GEN) was administered intravenously with a cocktail renal marker.Through a comprehensive analysis of the protein expression in the transporter related to tubular secretion and the concentration of substrates in serum and urine at different periods,it was shown that the glomerular filtration function of rats was compensated and the tubular secretion function decreased slightly in 25 and 50 mg/(kgd tubule secretion ·day) GEN groups.With the aggravation of injury,both the glomerular filtration anwere decreased obviously.The area under the concentration-time curve of serum creatinine,5-fluorouracil,furosemide,metformin and p-aminohippurate increased in 75 and 100 mg/(kg·day) GEN groups,and the clearance rate of them decreased in varying degrees except for furosemide.
