摘 要:目的研究多囊卵巢综合征(PCOS)患者外周血中环状RNA(circRNA)的表达差异,探讨circRNA在PCOS发病中作用机制。方法收集2021年6—8月中国科学大学深圳医院(光明)妇科就诊的3名PCOS患者和3名正常对照者的血液样本和常规资料,利用高通量测序技术检测样本中circRNA的表达差异,对筛选出的差异表达circRNA进行GO、KEGG基因富集分析。结果高通量测序共检测到23814种可分析的环状RNA,其中表达上调23个,表达下调的7个;差异表达RNA亲本基因功能富集分析显示,PCOS患者体内生物过程、细胞成分、分子功能等生物学功能都发生显著的变化。结论多种环状RNA分子在多囊卵巢综合征(PCOS)患者外周血中呈现差异表达;差异表达的环状RNA分子对PCOS患者生物学功能有一定的影响,可能参与PCOS的发病过程。Objective To explore the mechanism of circular RNA(circRNA) in the pathogenesis of polycystic ovary syndrome(PCOS), the expression difference of circRNA in the peripheral blood of patients with PCOS was studied. Methods The blood samples and routine data of 3 PCOS patients and 3 normal controls who visited the Department of Gynecology,University of Chinese Academy of Sciences Shenzhen Hospital in June to August 2021 were collected, and the expression differences of circRNA in the samples were detected by high-throughput sequencing technology. The parental genes expressing circRNA were subjected to GO and KEGG enrichment analysis. Results A total of 23814 analyzable circRNAs were detected by high-throughput sequencing, of which 23 were up-regulated and 7 were down-regulated. The functional enrichment analysis of the parental genes of differentially expressed RNA showed that biological processes, cellular components, and molecules in PCOS patients significant changes have taken place in biological functions such as functions. Conclusion A variety of circular RNA molecules are differentially expressed in the peripheral blood of patients with polycystic ovary syndrome.Differentially expressed circRNA molecules have a certain impact on the biological functions of PCOS patients, and may be involved in the pathogenesis of PCOS.
