摘  要:目的:探索石斛合剂对2型糖尿病合并非酒精性脂肪肝小鼠模型的效应机制。方法:将db/db雄性小鼠随机分为模型组、石斛合剂低剂量组、石斛合剂中剂量组、石斛合剂高剂量组,用低蛋氨酸低胆碱饲料喂养4周,制备T2DM合并NASH模型,将同窝出生的db/m小鼠设为正常组。正常组、模型组给予蒸馏水(0.1mL/10g)灌胃,石斛合剂低剂量组给予1/2SHHJ临床等效量灌胃(0.1mL/10g),石斛合剂中剂量组给予SHHJ临床等效量灌胃(0.1mL/10g),石斛合剂高剂量组给予2*SHHJ临床等效量灌胃(0.1mL/10g),连续灌胃4周。用HE、Masson染色处理肝脏组织切片,光镜观察病理学改变;Western Blot法检测小鼠肝脏中mTOR、TFEB、NF-κB、IL-18、IL-1β、TNF-α蛋白的表达。结果:在HE、Masson染色实验中,与模型组小鼠比较,石斛合剂各组小鼠肝脏的脂肪空泡和炎性细胞浸润均减轻,纤维化减少。在Western Blot实验中,与模型组比较,小鼠肝脏中mTOR、NF-κB、IL-18、IL-1β、TNF-α的水平降低(P<0.05),TFEB表达增加(P<0.05),其中均以石斛合剂中剂量组改善最为明显,其次为石斛合剂高剂量组。结论:石斛合剂对T2DM合并NASH有改善作用,其中石斛合剂中剂量疗效最为明显,其作用可能与mTOR-TFEB-NF-κB通路有关。Objective:To explore the effect mechanism of SHHJ on type 2 diabetes with non-alcoholic steatohepatitis in mice.Methods:The db/db male mice were randomLy divided into model group,SHHJ low-dose group,SHHJ mediumdose group,SHHJ high-dose group,fed with methionine and choline deficient diet feed for 4weeks to prepare T2DM combined with NASH model.db/m mice born in the same litter were used as the normal group,The normal group and the model group were given distilled water(0.1mL/10g)by gavage,the SHHJ low-dose group was given 1/2SHHJ clinically equivalent gavage(0.1mL/10g),the SHHJ middle-dose group was given SHHJ clinically equivalent gavage(0.1mL/10g),the SHHJ high-dose group was given 2*SHHJ clinically gavage(0.1mL/10g),equivalent gavage for 4weeks.Hematoxylin-eosin(HE)staining and Masson staining were used to process liver tissue sections,and pathological changes were observed under light microscope;Western Blot method was used to detect the expression of mTOR,TFEB,NF-κB,IL-18,IL-1β,TNF-αin the mouse liver.Results:In the HE and Masson staining experiments,compared with the model group of mice,the fatty vacuoles,the inflammatory cell infiltration and the fibrosis was reduced in the liver of the SHHJ groups were all reduced.In the Western Blot experiment,compared with the model group,the levels of mTOR,NF-κB,IL-18,IL-1β,TNF-αin the liver of mice decreased(P<0.05),and the expression of TFEB increased(P<0.05),among all which the SHHJ middle-dose group improved the most,and followed by the SHHJ high-dose group.Conclusion:SHHJ can improve the situation of T2DM complicated with NASH,and the SHHJ middle-dose group has the most obvious effect,which may be related to the mTOR-TFEB-NF-κB pathway.
