摘 要:目的分析酶联免疫分析法检测乙型肝炎表面抗原(HBsAg)常见影响因素。方法回顾性分析汝阳县中医院2019年10月至2021年5月接收的慢性乙型肝炎患者44例血清样本作为研究对象。抽取患者肘静脉血5 mL,离心处理后留下血清,将44份血清平均分成四等份,计为A、B、C、D,使用酶联免疫分析法检测HBsAg。比较不同时间HBsAg阳性血清样本滴度OD值、不同温度HBsAg阳性血清样本滴度OD值、不同洗扳HBsAg阳性血清样本滴度OD值和三种不同酶标记抗体加入量HBsAg阳性血清样本滴度OD值。结果不同时间点5 min、10 min、15 min、20 min、25 min、30 min、35 min、40 min平均OD值比较差异有统计学意义(P<0.05)。其中40 min平均OD值(0.71±0.02)最高,5 min平均OD值(0.41±0.02)最低。37℃下HBsAg阳性血清样本滴度OD值高于12℃(P<0.05)。自来水HBsAg阳性血清样本OD值高于试剂盒洗液(P<0.05)。40μL、50μL、60μL酶标记抗体加入量的OD值比较差异有统计学意义(P<0.05),其中50μL酶标记抗体加入量OD值(0.77±0.19)最高,40μL酶标记抗体加入量OD值(0.72±0.12)最低。结论时间、洗板、温度是酶联免疫分析法检测乙肝表面抗原常见影响因素,要准确处理。【Objective】To investigate the common factors influencing the detection of hepatitis B surface antigen(HBsAg)by enzyme-linked immunoassay.【Methods】Serum samples of 44 patients with chronic hepatitis B admitted in our hospital from October 2019 to May 2021 were collected.After centrifugation,44 serum samples were equally divided into four sets,and labeled as A,B,C and D.Enzyme-linked immunoassay was used for HBsAg.The average OD value of HBsAg positive serum samples at different time,different temperature,different eluent and different enzyme-labeled antibodies were compared.【Results】The average OD values of 5 min,10 min,15 min,20 min,25 min,30 min,35 min and 40 min showed significant difference(P<0.05).The average OD value at 40 min was the highest(0.71±0.02),and that at 5 min was the lowest(0.41±0.02).The OD value of HBsAg positive serum samples at 37°C was higher than that at 12°C(P<0.05).The OD value of HBsAg-positive serum samples in tap-water was higher than that of washing liquid(P<0.05).OD value of 40μL,50μL,and 60μL enzyme-labeled antibody was showed significant difference(P<0.05).The OD value of 50μL enzyme-labeled antibody was the highest(0.77±0.19),and that of 40μL enzyme-labeled antibody was the lowest(0.72±0.12).【Conclusion】Time,eluent and temperature are the common influencing factors of ELISA in detecting HBsAg,which should be dealt with accurately.
