摘 要:目的:观察芍药苷对慢性盆腔炎(CPID)大鼠抗纤维化和抗炎的治疗作用,探究其作用机制。方法:取50只未孕雌鼠经苯酚胶浆法建立大鼠CPID模型,另取10只大鼠作为假手术组,2周后将造模大鼠随机分为模型组、地塞米松组(2 mg/kg)和芍药苷低、中、高剂量组(25、50、100 mg/kg),连续治疗30 d。观察大鼠的一般状态,计算子宫系数,ELISA法检测肿瘤坏死因子-α(TNF-α)、C反应蛋白-1(CRP-1)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的含量,HE染色和Masson染色观察大鼠子宫病变情况,Western Blot法检测子宫组织转化生长因子-β1(TGF-β1)、Smad2/3、p-Smad2/3、Smad7蛋白表达。结果:与假手术组比较,CPID模型大鼠生存状态较差,子宫系数升高,血清炎症因子TNF-α、CRP-1、IL-1β、IL-6的水平升高,子宫宫腔可见有明显缩窄,炎症和纤维化程度明显,TGF-β1、p-Smad2/3的表达升高,Smad7的表达降低(P<0.05)。与模型组比较,芍药苷改善了大鼠生存状态,降低了子宫系数,降低血清炎症因子水平,改善子宫组织炎症和纤维化病变,降低TGF-β1、p-Smad2/3的表达,提高了Smad7的表达水平(P<0.05)。结论:芍药苷对CPID大鼠的纤维化和炎症状态具有治疗作用,其机制可能与抑制TGF-β/Smads通路的激活有关。Objective:To observe the anti-fibrosis and anti-inflammatory therapeutic effects of paeoniflo⁃rin on chronic pelvic inflammatory disease(CPID)in rats,and to explore its action mechanism.Methods:50 non-pregnant female rats were established rat model of CPID by the phenol glue method,and another 10 rats were assigned into the sham operation group.Two weeks later,the successful model rats were randomly divided into the model group,the dexamethasone group(2 mg/kg),the paeoniflorin groups of low-dose(25 mg/kg),medium-dose(50 mg/kg)and high-dose(100 mg/kg).After 30 continuous treatment,the general state of the rats was observed;uterine coefficient was calculated;the contents of tumor necrosis factor-α(TNF-α),C-reactive protein-1(CRP-1),interleukin-1β(IL-1β)and interleukin-6(IL-6)were detected by ELISA;the uterine lesions in rats were observed by HE staining and Masson staining;Western Blot was used to detect the protein expressions of transforming growth factor-β1(TGF-β1),Smad2/3,p-Smad2/3 and Smad7 in uterine tissues.Results:Compared with those in the sham operation group,the survival status of the rats was worse,the uterine coefficient was increased,the serum levels of TNF-α,CRP-1,IL-1βand IL-6 were increased.The uterine cavity was obviously narrowed,inflamma⁃tion and fibrosis were obvious,the expressions of TGF-β1 and p-Smad2/3 was increased,and the expres⁃sion of Smad7 was decreased in the model group(P<0.05).Compared with those in the model group,the survival status of the rats was improved,the uterine coefficient was reduced,the serum levels of inflammatory factors were lowered,the uterine tissue inflammation and fibrosis were improved,the expressions of TGF-β1 and p-Smad2/3 were decreased,and the expression level of Smad7 was increased in the Paeoniflorin groups(P<0.05).Conclusion:Paeoniflorin has a therapeutic effect on anti-fibrosis and anti-inflammation in CPID rats,and its mechanism may be related to inhibiting the activation of TGF-β/Smads pathway.
