摘  要:针对钼损伤绵羊瘤胃组织影响消化功能的问题,选取20只健康小尾寒羊[(20.61±1.50)kg]随机分为5组,钼处理组绵羊按体质量分别经口灌喂(5 mg/kg、10 mg/kg、20 mg/kg和50 mg/kg)钼酸钠,对照组灌喂等量蒸馏水。钼处理第28天时,用胃管式瘤胃液采样器采集瘤胃液。然后处死绵羊,采集瘤胃腹囊部上皮组织,用于检测瘤胃液消化酶及瘤胃上皮组织病理形态学变化研究。研究结果显示:对照组和钼5组瘤胃上皮细胞排列紧密,钼10组、钼20组和钼50组瘤胃上皮细胞间隙增大,细胞核淡染,染色质丢失,其中钼50组损伤最为严重。对照组瘤胃上皮细胞线粒体双层膜和嵴清晰可见,钼5组瘤胃上皮细胞线粒体嵴轻度肿胀,10 mg/kg、20 mg/kg和50 mg/kg的钼酸钠添加导致瘤胃上皮细胞线粒体嵴断裂甚至溶解,线粒体基粒数目明显减少。与对照组相比,钼5组、钼10组和钼20组纤维素酶活性显著上升(P<0.05或P<0.01),钼10组、钼20组和钼50组α-淀粉酶活性和乳酸浓度极显著上升(P<0.01)。高剂量的钼造成瘤胃上皮病理性损伤以及消化酶活性的改变。Aim at the problem that molybdenum(Mo)damages rumen tissue and effects digestive function of sheep,in this paper,twenty healthy small-tailed Han sheep[(20.61±1.50)kg]were randomly divided into five groups.The sheep in Mo treatment groups were orally fed with sodium molybdate(5 mg/kg,10 mg/kg,20 mg/kg and 50 mg/kg)according body weight,while control group were fed with same amount of distilled water.On the 28th day of Mo treatment,rumen fluid was collected by gastric tube rumen fluid sampler.Then the sheep were sacrificed to collect the rumen abdominal sac epithelial tissue,which were used to detect the changes of digestive enzymes in rumen fluid and histopathological changes of rumen epithelium.The results showed that rumen epithelial cells in the control group and Mo 5 group are closely arranged.However,rumen epithelial cell space is enlarged,nucleus is lightly stained,and chromatin is lost in the Mo 10 group,Mo 20 group and Mo 50 group.The damage is most serious in Mo 50 group.The mitochondrial bilayer membrane and cristae of rumen epithelial cells are clearly visible in the control group,while mitochondrial cristae are slightly swollen in Mo 5 group.The addition of 10 mg/kg,20 mg/kg and 50 mg/kg sodium molybdate causes the mitochondrial cristae fracture and even dissolution.The number of mitochondrial grana obviously is decreased.Compared with the control group,cellulase activity in Mo 5 group,Mo 10 group and Mo 20 group significantly is increased(P<0.05 or P<0.01),andα-amylase activity(P<0.01)and lactic acid concentration(P<0.01)in Mo 10 group,Mo 20 group and Mo 50 group are extremely significant increased.High dose Mo causes pathological damage of rumen epithelium and changes of digestive enzymes activity.
