摘  要:目的代谢相关性脂肪肝(MAFLD)以肝细胞脂肪沉积和血脂异常为特征。文章旨在基于代谢组学探讨高脂诱导的MAFLD模型大鼠血清脂质代谢物特点。方法12只SD雄性大鼠随机数字表法分为正常组和模型组,每组6只,分别予普通饲料和高脂饲料喂养4周。油红O染色观察两组大鼠肝脏脂质沉积情况,利用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(UHPLC-QE-MS)非靶标代谢组学技术,筛选出潜在的差异代谢物并进行脂质富集分析。结果油红O染色结果显示模型组肝细胞内出现大量大小不等的红色脂滴堆积。代谢组学筛选出85个潜在差异代谢物,43个脂质差异代谢物,7种脂类物质,其中脂肪酸类、固醇脂类和甘油磷脂类差异有统计学意义(P<0.05)。结论MAFLD模型大鼠血清脂质代谢物存在显著变化。Objective Metabolic associated fatty liver disease(MAFLD)is the most common chronic liver disease in the world and is characterized by fatty deposition in hepatocytes and dyslipidemia.The aim of this study was to investigate the characteristics of serum lipid metabolites in high-lipid-induced MAFLD model rats based on metabonomics.Methods 12 male SD rats were divided into normal group and model group by random number table method,with 6 rats in each group,and fed with normal diet and high-fat diet respectively for 4 weeks.Oil red O staining was used to observe the lipid deposition in the liver of rats in the two groups.The potential differential metabolites were screened out and the lipid enrichment was analyzed by non-target metabolomics technique of ultra-high performance liquid chromatography-quadrupole-electrostatic field track well high resolution mass spectrometry(UHPLC-QE-MS).Results Oil red O staining results showed that a large number of red lipid droplets with different sizes were accumulated in the liver cells of model group.Metabolomics screened 85 potential differential metabolites,43 lipid differential metabolites and 7 lipid substances,among which fatty acids,sterols and glycerophospholipids had statistical significance(P<0.05).Conclusion Serum lipid metabolites of MAFLD model rats were significantly changed.
