针刺结合蹊径通脉汤对心肌梗死大鼠ATP敏感性钾离子通道相关蛋白Kir6.1、Kir6.2的影响

摘　　要：目的:以“针药结合对心肌梗死(MI)大鼠ATP敏感性钾离子通道相关蛋白Kir6.1、Kir6.2表达的影响”为研究目的,研究针药结合对心肌梗死改善的可能机制,为实现研发心肌梗死新治疗手段提供实验数据基础。方法:65只健康雄性SD大鼠,随机选择10只作为对照组。其余大鼠饲以高脂肪食物,共3周,对照组大鼠皮下多点注射生理盐水,其余大鼠按相同方式注射盐酸异丙肾上腺素。通过心电图对比,将造模成功的40只MI大鼠随机分为模型组、针刺组、西药组、针药结合组,每组10只。分别给予相应治疗后,观察各组大鼠心电图变化,用HE染色观察大鼠心肌细胞的病理改变,以Western blot检测技术检测ATP敏感性钾离子通道(Kir6.1、Kir6.2)蛋白的表达量。结果:与对照组相比,实验组的40份实验标本在心肌细胞蛋白(Kir6.1、Kir6.2)的表达量上都有所提高,差异具有统计学意义(P<0.05)。治疗后,与模型组相比,西药组、针刺组、针药结合组的心肌细胞Kir6.1、Kir6.2的蛋白表达量均呈下降趋势,其中针药结合组下降程度最明显,差异具有统计学意义(P<0.05)。针刺组和西药组的下降程度不大,差异无统计学意义(P>0.05)。结论:针药结合对改善心肌梗死大鼠疗效显著,可能与大鼠心肌细胞中ATP敏感性钾离子通道相关蛋白Kir6.1、Kir6.2的表达有关。Objective:To study the effect of acupuncture and drug combination on the expression of ATP-sensitive potassium ion channel-related proteins Kir6.1 and Kir6.2 in rats with myocardial infarction,to explore the possible mechanism of acupuncture and drug combination on the improvement of myocardial infarction,and to provide experimental data basis for the development of new treatment methods for myocardial infarction.Methods:Sixty-five healthy male SD rats were randomly selected as control group. The other rats were fed a high-fat diet for three weeks. The control rats were subcutaneously injected with multiple points of normal saline,and the other rats were injected with isoproterenol hydrochloride in the same way. By electrocardiogram comparison,40 MI rats successfully modeled were randomly divided into model group,acupuncture group,Western medicine group and acupuncture and drug combination group,with 10 rats in each group. After corresponding treatment,ecg changes of rats in each group were observed,pathological changes of cardiac myocytes were observed by HE staining,and protein expression levels of ATP sensitive potassium channels(Kir6.1 and Kir6.2)were detected by Western blot.Results:Compared with the control group,the protein expression level of myocardial cells in 40 experimental specimens in the experimental group was increased,which had experimental value(P<0.05). After treatment,compared with model group,the protein expression levels of Kir6.1and Kir6.2 in cardiomyocytes of western medicine group,acupuncture group and acupuncture and drug combination group showed a downward trend,and the decrease degree of acupuncture and drug combination group was the most obvious,which had experimental value(P<0.05). There was no significant decrease in acupuncture group and western medicine group(P>0.05).Conclusion:Combination of acupuncture and drug has significant effect on improving myocardial infarction in rats,which may be related to the expression of ATP sensitive potassium channel related proteins Kir6.1 and Ki
