加味九分散对痛风性关节炎大鼠TRPM8表达的影响

摘　　要：目的以痛风性关节炎大鼠为研究对象,通过外敷加味九分散观察大鼠关节肿胀情况并探讨其止痛机制。方法将60只SD大鼠随机分为空白对照组、模型组、扶他林乳胶剂组、加味九分散外敷组4组,除空白组以外,模型组、扶他林乳胶剂组、加味九分散外敷组均制造大鼠痛风性关节炎模型,同时扶他林乳胶剂组和加味九分散组给药48 h后取大鼠关节囊及关节周围皮肤提取蛋白,通过Western blot测定TRPM8的蛋白表达情况,ELISA测定血清中白细胞介素6(IL-6)含量。实验数据通过SPSS23软件进行分析。结果扶他林乳胶剂组和加味九分散组增加了大鼠TRPM8的相对表达量,扶他林乳胶剂组和加味九分散组中IL-6表达量下调,与模型组比较差异有统计学意义(P<0.01)。结论加味九分散可能是通过增加TRPM8的表达,下调IL-6起到止痛作用。Objective To take gouty arthritis rats as the research object,observe the joint swelling of rats by external application of Jiawei Jiufen Powder(加味九分散)and explore its analgesic mechanism.Methods 60 SD rats were randomly divided into four experimental groups:blank control group,model group,Futalin emulsion group and Jiawei Jiufen Powder group.Excepted for blank group,model group,Futalin latex group and Jiawei Jiufen Powder group were all made models of gouty arthritis in rats.At the same time,after 48 hours of administration,the articular capsule and the skin around the joint of rats in the Futalin emulsion combination group were taken to extract proteins.The protein expression of TRPM8 was determined by Western blot,and the serum interleukin 6(IL-6)content was determined by ELISA.The experimental data were analyzed by SPSS23 software.Results The relative expression level of TRPM8 was increased in Futarin emulsion group and Jiawei Jiufen Powder group,and the expression level of IL-6 was down-regulated in Futarin emulsion group and Jiawei Jiufen Powder group,which was significantly different from that in model group(P<0.01).Conclusion Jiawei Jiufen Powder may play an analgesic role by increasing the expression of TRPM8 and down-regulating IL-6.
