摘 要:目的通过多指标综合评分法优选智脑胶囊最佳醇提工艺。方法采用L9(34)正交设计方法,考察乙醇浓度、溶剂倍数、回流时间和提取次数4个因素,每因素设置3个水平;以松果菊苷、毛蕊异黄酮苷、党参炔苷、槲皮素、多糖含量及浸膏得率为指标进行考察。采用UPLC法测定指标成分含量,色谱条件:色谱柱为ACE C18柱(100 mm×2.1 mm),流动相为乙腈(A)和0.1%甲酸溶液(B),梯度洗脱,检测波长为260 nm,进样体积2μL;采用苯酚-硫酸法于490 nm波长处测定多糖含量;采用热浸法测定浸膏含量。结果最佳醇提取工艺:75%浓度的乙醇为提取溶剂,第1次加10倍量,回流提取60 min,第2次加8倍量,回流提取40 min;在此条件下得到的4种指标成分和多糖平均含量分别为5.612、0.544、0.427、0.188、12.47 mg/g,浸膏得率为22.37%。结论通过多指标综合加权评分法优选出的最佳醇提工艺操作简便、稳定可行、重复性好,为实际生产提供了参考。Objective The optimum ethanol extraction process of Zhinao Capsule(智脑胶囊)was optimized by multi-index comprehensive scoring method.Methods L9(34)orthogonal design method was used to investigate the ethanol concentration,solvent multiple,reflux time and extraction times,and three levels were set for each factor.The contents of echinacea glycoside,calycosin isoflavone glycoside,codonopsis acetylene glycoside,quercetin,polysaccharide and extract yield were investigated as indexes.The chromatographic conditions were as follows:ACE C18 column(100 mm×2.1 mm),acetonitrile(A)and 0.1% formic acid solution as the mobile phase(B),gradient elution,260 nm was set as detection wavelength,and sample volume was 2μL.The content of polysaccharide was determined by phenolsulfuric acid method at 490 nm wavelength.The content of extract was determined by hot dip method.Results The optimum extraction conditions were as follows:The best ethanol extraction process was 75% ethanol as the extraction solvent,10 times the first time,60 minutes reflux,8 times the second time,and 40 minutes.Under these conditions,the average contents of four index components and polysaccharides were 5.612 mg/g,0.544 mg/g,0.427 mg/g,0.188 mg/g and 12.47 mg/g,respectively,and the extract yield was 22.37%.Conclusion The optimum ethanol extraction process is simple,stable,feasible and reproducible,which provides a reference for actual production.
