振腹环揉法对PCPA失眠模型大鼠脑电活动及下丘脑5-HT、β-EP含量的影响

摘　　要：目的通过实验动物脑电实验以及血清酶联免疫检测,观察振腹环揉法对PCPA(对氯苯丙氨酸)失眠模型大鼠脑电活动及下丘脑5-HT、β-EP含量的影响。方法选择6月龄SD大鼠18只(雌雄各半),在电极埋置术后1周随机分为空白组、模型组、推拿组(n=6)。对模型组和推拿组大鼠进行PCPA法造模。造模成功后,将模型组大鼠于束缚鼠板仰卧位每日1次;对推拿组大鼠进行“振腹环揉法”腹部推拿治疗每日1次,连续7 d;7 d后,分别对各组大鼠进行脑电监测和血清酶联免疫检测。结果模型组与空白组相比,δ波百分比降低,差异具有统计学意义(P<0.001);θ波、α波、β波百分比升高,差异具有统计学意义(P<0.05);下丘脑内5-HT、β-EP含量下降,差异具有统计学意义(P<0.001)。推拿组与模型组相比,δ波百分比升高,差异具有统计学意义(P<0.05);θ波百分比降低,差异具有统计学意义(P<0.05)。下丘脑内5-HT的含量升高,差异具有统计学意义(P<0.05);β-EP的含量升高,差异具有统计学意义(P<0.001)。结论振腹环揉法对PCPA失眠模型大鼠脑电活动及下丘脑5-HT、β-EP含量有影响作用。Objective To observe the effects of the abdominal massage on the electroencephalogram(EEG)activities and the contents of 5-HT andβ-EP in the hypothalamus of the paca-chlorophenylalanine(PCPA)induced model rats with insomnia through the electroencephalography experiment and serum enzyme-linked immunosorbent assay.Methods A total of eighteen 6-month-old SD rats(half male and half female)were selected and randomly divided into a blank group,a model group and a massage group(n=6)one week after the electrode implantation.The rats in the model group and massage group were modeled with PCPA.After modeling,the rats in the model group were placed in the supine position on the restraint plate once a day,while those in the massage group were treated with the abdominal massage once a day,both for 7 consecutive days.After that,an EEG monitoring and a serum enzyme-linked immunosorbent assay were performed on the rats in each group.Results Compared with the blank group,the percentage ofδwave in the model group was significantly decreased(P<0.001),while the percentages ofθwave,αwave andβwave were significantly increased(P<0.05).The contents of 5-HT andβ-EP in hypothalamus were significantly decreased(P<0.001).Compared with the model group,the percentage ofδwave in the massage group was significantly increased(P<0.05),while the percentage ofθwave was significantly decreased(P<0.05).The content of 5-HT in the hypothalamus was significantly increased(P<0.05),while the content ofβ-EP was significantly increased(P<0.001).Conclusion The abdominal massage has significant effects on the EGG activities and the contents of 5-HT andβ-EP in the hypothalamus of the PCPA model rats with insomnia.
