摘 要:目的探讨联氨基姜黄素(HZC)对二乙基亚硝胺(DEN)诱导雄性SD大鼠肝癌发生、发展的影响。方法根据实验设计将选取的80只SD雄性大鼠随机分为正常对照组(10只)、药物对照组(10只)、模型组(30只)、干预组(30只)。正常对照组以80 mg/kg给予生理盐水腹腔注射;药物对照组以80 mg/kg给予HZC腹腔注射;模型组以50 mg/kg给予DEN腹腔注射;干预组以50 mg/kg DEN和80 mg/kg HZC腹腔注射,以上4组给药方法均2次/w,12 w后停药。待24 w实验结束,利用组织学、免疫组化和组织生化等检测方法,观察HZC对DEN诱发大鼠肝癌的影响。结果正常对照组、药物对照组肝组织均未发生癌变。与模型组相比,干预组肝癌发病率低、存活率高、肝脏癌结节小而少,差异具有统计学意义(P<0.05)。干预组体重、肝重、肝指数与模型组比较差异显著(P<0.01)。两个对照组均具有正常肝小叶结构,模型组显示出癌灶;干预组显示癌灶伴片状坏死。免疫组化显示两个对照组无增殖细胞核抗原(PCNA)染色,干预组肝组织PCNA阳性率明显低于模型组(P<0.01)。肝组织血浆中谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、γ-谷氨酰转移酶(GGT)、总胆红素水平(TBL)也显著低于模型组(P<0.05)。结论HZC抑制了DEN诱发大鼠肝癌的进程,并具有防护作用,对降低肝癌发生和减少DEN对大鼠肝脏的毒性作用具有显著效果。Objective To investigate the influence of hydrazinocurcumin(HZC)on the occurrence and development of SD rat hepatocarcinoma induced by diethylnitrosamine(DEN).Methods A total of 80 healthy male SD rats were randomised divided into four groups accordance with the requirements:normal control group(n=10),drug control group(n=10),model group(n=30),intervention group(n=30).Normal control group was administered intraperitoneally physiological saline(80 mg/kg);drug control group was administered intraperitoneally HZC(80 mg/kg);model group was given intraperitoneal injection of DEN(50 mg/kg);intervention group were given intraperitoneal injection of DEN(50 mg/kg)and HZC(80 mg/kg).The above four groups were administered twice a week for 12 weeks.At the end of 24 weeks,histology,immunohistochemistry,serum and tissue biochemistry methods were used to study the effect of HZC on the effect of DEN induced hepatocarcinoma of rats.Results No canceration of liver tissue were observed in normal control group and drug control group.Compared with the model group,the incidence of liver cancer in the intervention group was lower,the survival rate was higher,and the nodules of liver cancer were smaller and less,the differences were statistically significant(P<0.05).There were significant differences in body weight,liver weight and liver index between the intervention group and the model group(P<0.05).The two control groups all had normal liver lobular structure.The model group showed cancerous lesions;the intervention group showed cancerous lesions with patchy necrosis.There was no PCNA staining in the two control groups.PCNA positive rate of liver tissue in the intervention group was significantly lower than that in the model group(P<0.01).The content of serum ALT,AST,ALP,GGT and TBL levels in the intervention group were significantly lower than those in the model group(P<0.05).Conclusions HZC could inhibit the process of DEN-induced hepatocarcinoma in rats and had a protective effect,it has a significant effect on decreasing the occu
