摘 要:目的:探讨黄芪甲苷联合葛根素对高糖诱导的H9c2细胞内质网应激及其凋亡相关因子的影响。方法:将对数生长期的H9c2细胞随机分为对照组、甘露醇对照组、高糖组、高糖+4-苯基丁酸(4-phenylbutyric acid,4-PBA)组、毒胡萝卜素(thapsigargin,TG)组、高糖+黄芪甲苷+葛根素组;CCK-8法检测H9c2细胞活力;Western blot检测凋亡相关蛋白B淋巴细胞瘤2(B-cell lymphoma-2,Bcl-2)和Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)表达。RT-qPCR及Western blot检测葡萄糖调节蛋白78(glucose-regulated protein 78,GRP78)、肌醇酶1α(inositol-re‐quiring enzyme 1α,IRE1α)、调控X盒结合蛋白1(X-box binding protein 1,XBP1)、转录因子C/EBP同源蛋白(C/EBP homologous protein,CHOP)、p53上调凋亡调节因子(p53 up-regulate modulator of apoptosis,PUMA)mRNA和蛋白的表达。结果:与对照组相比,高糖组Bax/Bcl-2比值显著增加(P<0.01),高糖组和TG组GRP78、IRE1α、XBP1、CHOP和PUMA表达增加(P<0.05),4-PBA或黄芪甲苷联合葛根素均可降低高糖诱导的GRP78、IRE1α、XBP1、CHOP和PUMA的高表达和Bax/Bcl-2比值(P<0.05)。结论:黄芪甲苷联合葛根素对高糖诱导的H9c2细胞内质网应激及凋亡相关因子有抑制作用。AIM:To investigate the effect of astragaloside Ⅳ(AS-Ⅳ)combined with puerarin on high glucose(HG)-induced endoplasmic reticulum stress(ERS) and apoptosis-related factors in H9c2 rat cardiomyocytes.METHODS:The H9c2 cells in logarithmic growth phase were randomly divided into control group,mannitol control group,high glucose(HG)group,HG+4-phenylbutyric acid(4-PBA)group,thapsigargin(TG)group,and HG+AS-Ⅳ+puerarin group. Cell viability was measured by CCK-8 assay. The expression of Bcl-2 and Bax was detected by Western blot. The mRNA and protein levels of glucose-regulated protein 78(GRP78),inositol-requiring enzyme 1α(IRE1α),Xbox binding protein 1(XBP1),C/EBP homologous protein(CHOP),and p53 up-regulated modulator of apoptosis(PUMA)were examined by RT-qPCR and Western blot.RESULTS:Compared with control group,the Bax/Bcl-2 ratio was significantly increased(P<0. 01)in the HG group. The mRNA and protein levels of GRP78,IRE1α,XBP1,CHOP,and PUMA were increased in the HG and TG group(P<0. 05). High glucose-induced expressions of the above factors were decreased in HG+4-PBA group(P<0. 05). Astragaloside Ⅳ combined with puerarin decreased the mRNA and protein levels of endoplasmic reticulum stress-related factors such as GRP78,IRE1α,XBP1,CHOP,and PUMA as well as the Bax/Bcl-2 ratio in high glucose-induced H9c2 cells(P<0. 05).CONCLUSION:Astragaloside Ⅳ combined with puerarin decrease high glucose-induced endoplasmic reticulum stress and apoptosis-related factor expression in H9c2 cells.
