DJ-1、miR-20b-5p在子宫内膜异位症中的表达及与抑癌基因PTEN的关系

摘　　要：目的:探讨DJ-1、miR-20b-5p在子宫内膜异位症(EMT)中的表达及与抑癌基因PTEN的关系。方法:选择2019年3月-2021年3月本院收治的行腹腔镜手术治疗65例EMT患者的在位内膜(Eu组)、异位内膜(Ec组)及行子宫肌瘤剔除术且排除EMT 60例正常内膜组织(对照组)。采用免疫组化法检测3组组织中DJ-1、PTEN蛋白表达水平,采用实时荧光定量PCR法检测3组组织中DJ-1、miR-20b-5p、PTEN mRNA表达水平。Pearson相关分析EMT组织中DJ-1、miR-20b-5p与PTEN mRNA表达关系。结果:免疫组化结果显示,对照组、Eu组、Ec组DJ-1蛋白表达水平依次升高,PTEN蛋白表达水平依次降低(均P<0.05)。PCR结果显示,对照组、Eu组、Ec组DJ-1、miR-20b?5p mRNA表达水平依次升高,PTEN mRNA表达水平依次降低(均P<0.05)。Pearson相关分析显示,DJ-1、miR-20b-5p mRNA与PTEN mRNA表达水平均呈负相关(r=-0.716、-0.587,均P<0.01)。结论:DJ-1、miR-20b-5p在EMT中表达上调,可能通过负性调节PTEN表达水平,增强异位内膜细胞增殖能力,从而导致EMT发生。Objective:To explore the expressions of DJ-1and miR-20b-5p in endometriosis(EMT)tissue,and to study the correlation between the DJ-1and miR-20b-5p levels and the level of tumor suppressor gene PTEN.Methods:The tissues of eutopic endometrium(in group A)and ectopic endometrium(in group B)from 65patients with EMT who had undergone laparoscopic surgery were selected in this study from March 2019to March 2021,and another normal endometrial tissues from 60patients who had undergone uterine myomectomy were selected in group C.The protein expression levels of DJ-1and PTEN in endometrial tissues of the three groups were detected by immunohistochemistry,and the expression levels of DJ-1,miR-20b-5p,and PTEN mRNA in endometrial tissues of the three groups were detected by real-time fluorescent quantitative PCR(qRT-PCR).Pearson correlation analysis was used to analyze the relationship between the levels of DJ-1and miR-20b-5p and the PTEN mRNA expression level in endometrial tissues of group A and B.Results:The results of immunohistochemistry showed that the expression level of DJ-1protein in group C,in group A,and in group B had increased gradually,while the expression level of PTEN protein in group C,in group A,and in group B had decreased gradually(all P<0.05).PCR results showed that the mRNA expression levels of DJ-1and Mir-20B-5p in group C,in group A,and in group B had increased gradually,while the mRNA expression level of PTEN in group C,in group A,and in group B had decreased gradually(P<0.05).Pearson correlation analysis showed that the expression levels of DJ-1and Mir-20B-5p mRNA were negatively correlated with the PTEN mRNA level(r=-0.716,-0.587,P<0.01).Conclusion:The expressions of DJ-1and miR-20b-5p in endometrial tissues of EMT are up-regulated,which lead to the occurrence of EMT by negatively regulating the expression of PTEN and enhancing the proliferation of ectopic endometrial cells.
