糠椴ATP合酶基因生物信息学分析

摘  要:ATP合酶是光合作用中光合磷酸化和呼吸作用氧化磷酸化的关键酶,可以催化合成ATP.利用生物信息学分析糠椴ATP合酶基因的同源性、开放阅读框、氨基酸组成、蛋白质亲/疏水性、蛋白质跨膜结构域、氨基酸二级结构,利用同源建模构建糠椴ATP合酶三维结构,MEGA7构建糠椴ATP合酶不同亚基基因系统进化树.结果表明:糠椴ATP合酶由atpA、atpB、atpE、atpF、atpH和atpI等6个亚基组成,每个亚基基因序列相似度达到100%的物种各不相同,且不同亚基的基因序列长度和氨基酸组成差异较大.其中,atpA、atpB、atpE和atpF为亲水性蛋白,atpH和atpI为疏水性蛋白.atpI的跨膜区最多,atpA、atpB和atpE不存在跨膜结构.糠椴ATP合酶由51.40%的α螺旋、13.10%的β折叠和35.50%的无规则卷曲组成,其中,atpF和atpH仅由α螺旋和无规则卷曲组成,无β折叠.糠椴atpA、atpB、atpE、atpF和atpI亚基基因与粉椴、少脉椴和紫椴亲缘关系较近.研究结果可为糠椴ATP合酶的基因克隆提供参考.ATP synthase is a key enzyme in photosynthesis and respiration,which can catalyze the synthesis of ATP.It makes bioinformatics analysis of ATP synthase gene in Tilia mandshurica.ATP synthase was composed of atpA,atpB,atpE,atpF,atpH and atpI subunits in T.mandshurica.The species with 100%similarity of gene sequence of each subunit were diverse,and the length of gene sequence and amino acid composition of different subunit were different.atpA,atpB,atpE and atpF were hydrophilic proteins,while atpH and atpI were hydrophobic proteins;atpI had the most transmembrane domain,and atpA,atpB and atpE had no transmembrane domain.ATP synthase was produced byα-helixs(51.40%),β-sheets(13.10%)and random coil(35.50%)in T.mandshurica.atpF and atpH were only composed ofα-helixs and random coil,and noβ-sheets.T.mandshurica,T.oliveri,T.paucicostata and T.amurensis in relationship were closer according to phylogenetic trees of atpA,atpB,atpE,atpF and atpI genes.These results can provide a reference for ATP synthase gene clone in T.mandshurica.
