摘 要:【目的】进一步研究菌草“绿洲一号”根际土壤中的微生物群落组成情况,以期筛选出其根际土壤中的有重要生态价值的优势菌株。【方法】通过对种植了菌草“绿洲一号”和未种植菌草“绿洲一号”的盐碱地分别进行土壤取样,运用16sDNA高通量测序对其进行微生物种群测序,明确两者之间的微生物种群差异,从而筛选出菌草“绿洲一号”根际土壤中的优势菌株,对有重要生态价值的优势菌群进行分离培养及菌落PCR鉴定。【结果】实验地段中的土壤微生物种群丰度要远远高于空白地段,同时实验地段中的土壤微生物种群结构组成相较于空白地段也具有明显变化,菌草“绿洲一号”根际土壤中有重要生态价值的优势菌株为酸杆菌和鞘氨醇单胞菌。对其进行分离培养之后,菌落PCR鉴定表明优势菌株被成功分离培养。【结论】本文通过对菌草“绿洲一号”根际土壤中具有重要生态价值的优势菌株进行筛选培养,为利用菌草“绿洲一号”改良盐碱土壤提供理论依据具有重要的指导意义。[Objective]The composition of microbial community in rhizosphere soil of Juncao‘Oasis No.1’was further studied in order to screen out dominant strains with important ecological value in rhizosphere soil.[Method]Soil samples were taken from saline and alkaline lands with and without Juncao‘Oasis No.1’,and microbial populations were sequenced using 16sDNA high-throughput sequencing to identify the differences between the two microbial populations,so as to screen out the dominant strains in rhizosphere soil of Juncao‘Oasis No.1’.The dominant bacteria with important ecological value were isolated,cultured and identified by Colony PCR.[Result]The abundance of soil microbial population in the experimental lot was much higher than that in the blank lot,and the composition of soil microbial population in the experimental lot also changed significantly compared with that in the blank lot,the abundance of ecologically important bacteria in the rhizosphere soil of Juncao‘Oasis No.1’was significantly increased.After isolation and culture,Colony PCR identification showed that the dominant strains were successfully isolated and cultured,which were Acidobacteria and Sphingomonas.[Conclusion]In this paper,the dominant strains with important ecological value in rhizosphere soil of Juncao‘Oasis No.1’were screened and cultivated,which provided theoretical basis for the improvement of salt-alkali soil by Juncao‘Oasis No.1’and had important guiding significance.
