摘  要:为探究云贵高原上植物的亲缘关系及地理分布格局,本研究通过对其18个居群个体材料的ITS、psbA-trnH和trnD-psbM序列进行了扩增以及测序,并结合杨梅属的杨梅和毛杨梅进行系统发育分析,探究云贵高原矮杨梅的亲缘关系及地理分布格局。结果显示:云贵高原矮杨梅ITS序列平均长度为657 bp,psbA-trnH序列平均长度为460 bp,trnD-psbM序列平均长度为603 bp,ITS+psbA-trnH+trnD-psbM联合序列总长度为1 720 bp,G+C平均含量为40.6%,共有58个变异位点和31个简约信息位点。基于ITS+psbA-trnH+trnD-psbM联合序列构建的系统发育树可看出,矮杨梅18个居群的单个序列与毛杨梅和杨梅区分开,单独聚为一支,支持矮杨梅为一单系类群。同时,系统发育树基本划分Ⅰ支系和Ⅱ支系,Ⅰ支系主要包含乌蒙山脉以东和南面的居群,而Ⅱ支系主要包含乌蒙山脉以西的居群,包括来自大理地区,云南高原中部和攀枝花地区的矮杨梅。亲缘关系较近的居群,其实际地理分布位置也较近,表明ITS+psbA-trnH+trnD-psbM联合序列可用于矮杨梅的亲缘关系和地理分布格局研究中。本研究为进一步探讨云贵高原植物区系演化及物种多样性提供重要的理论基础,为矮杨梅资源的收集、保存、分类鉴定和合理开发利用提供参考依据。In order to explore the phylogenetic relationship and geographical distribution pattern of plants on the Yunnan-Guizhou Plateau,this study amplified and sequenced ITS,psbA-trnH and trnD-psbM sequences from 18 populations of Myrica nana endemic to the Yunnan-Guizhou Plateau,and conducted phylogenetic analysis combining Myrica rubra and Myrica esculenta of the genus Myrica L.,and to explore the genetic relationship and geographical distribution pattern of Myrica nana in Yunnan-Guizhou Plateau.The results showed that the average length of ITS sequence was 657 bp,the average length of psbA-trnH sequence was 460 bp,the average length of trnD-psbM sequence was 603 bp,the total length of ITS+psbA-trnH+trnD-psbM combined sequence was 1 720 bp,the average G+C content was 40.6%,and there are a total of 58 variation sites and 31 reductive information sites.The phylogenetic tree based on ITS+psbA-trnH+trnD-psbM combined sequence showed that Myrica nana was a monophyletic branch,and the whole species was divided intoⅠbranch and Ⅱ branch with the Wumeng Mountain system as the boundary.TheⅠbranch mainly includes the populations to the east and south of Wumeng Mountain,while the Ⅱ branch mainly includes the populations to the west of Wumeng Mountain,including Dali area,central Yunnan Plateau and Panzhihua area.The actual geographical distribution of the populations with close genetic relationship was also close,indicating that the combined sequence of ITS+psbA-trnH+trnD-psbM could be used to study the genetic relationship and geographical distribution pattern of Myrica nana.This study provides an important theoretical basis for further study on the evolution of flora and species diversity in Yunnan-Guizhou Plateau.It provides reference basis for collection,preservation,classification and identification,rational development and utilization of Myrica nana.
