心舒宝片活血化瘀功效的有效物质辨识研究

摘　　要：目的:以含药肠吸收液-体外药理活性相结合的方法,初步辨识研究心舒宝片体现活血化瘀功效的有效物质。方法:制备心舒宝片全方及各单味药的肠吸收液,以抗凝血和抗氧化为反映活血化瘀的药理活性作用,寻找药效最强的单味药。抗凝血作用选择凝血4项和腺苷二磷酸(ADP)诱导血小板凝集实验,抗氧化作用选择1,1-二苯-2-三硝苯肼(DPPH)自由基清除实验。结果:抗凝血研究结果表明与空白对照组比较,山楂可延长凝血酶原时间(PT,P<0.01)和凝血酶时间(TT,P<0.05),且有较强的抗ADP诱导血小板凝集作用(P<0.01)。抗氧化研究结果表明与空白对照组比较,以丹参的DPPH自由基清除作用最强(P<0.01),其次分别为山楂、刺五加、白芍和郁金。结论:丹参和山楂可能是心舒宝片体现活血化瘀功效的重要组分,但尚不能代表全方的整体效应。Objective:To preliminarily study the therapeutic material basis of Xinshubao tablet in“promoting blood circulation and removing stasis”by a combined method of the drug-containing intestinal absorption liquid and the pharmacological activity evaluation in vitro.Methods:The intestinal absorption liquids containing Xinshubao prescription and five single ingredients were prepared,and the pharmacological effect of“promoting blood circulation and removing stasis”was reflected by the anticoagulant and antioxidant to find the single ingredient with the optimum efficacy.Four coagulation tests and adenosine diphosphate(ADP)-induced platelet aggregation test were conducted to detect the anticoagulant effect,and 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging test was conducted to reveal the antioxidant effect.Results:The results of anticoagulant experiments showed that compared with the blank control group,Crataegi Fructus prolonged prothrombin time(PT)(P<0.01)and thrombin time(TT)(P<0.05),and had a stronger effect on anti-platelet aggregation induced by ADP(P<0.01).The results of antioxidant experiments showed that compared with the blank control group,Salvia Root had the optimum effect on scavenging DPPH radical(P<0.01),followed by Crataegi Fructus,Acanthopanacis,Senticosi Radix Et Rhizoma Seu Caulis,Paeoniae Radix Alba,and Curcumae Radix.Conclusion:Salviae Miltiorrhizae Radix Et Rhizoma and Crataegi Fructus may be the most important ingredients of Xinshubao tablet to exert the pharmacological effect of“promoting blood circulation and removing stasis”,but they are unable to represent the whole effect of Xinshubao tablet.
