益肾活血丸调控Wnt7a/HOXA10改善内膜容受性的机理研究

摘　　要：目的:探讨益肾活血丸调控Wnt7a/同源盒基因10(homoboxA10,HOXA10)改善内膜容受性的作用机理。方法:SPF级雌性SD大鼠随机分为正常组、模型组、米非司酮组、益肾活血丸组及米非司酮+益肾活血丸组。模型组及益肾活血丸组大鼠复制子宫内膜容受性不良大鼠模型,米非司酮组及米非司酮+益肾活血丸组大鼠复制胚胎着床障碍动物模型。妊娠第1天开始,益肾活血丸组及米非司酮+益肾活血丸组大鼠以1.733 g·kg^(-1)·d^(-1)的剂量灌胃给予益肾活血丸溶液,其余组大鼠给予蒸馏水灌胃,每天1次,每次2 mL。实时荧光定量PCR检测子宫内膜组织中HOXA10 mRNA、Wnt7a mRNA的水平;Western Blot和免疫组织化学技术检测子宫内膜组织中HOXA10、Wnt7a的蛋白水平;并采用相关性分析考察HOXA10与Wnt7a的相关性。结果:与正常组比较,模型组、米非司酮组大鼠子宫内膜Wnt7a mRNA、HOXA10 mRNA的表达量显著下降(P<0.05),HOXA10、Wnt7a蛋白表达量及阳性单位(positive unit,PU)值均显著下降(P<0.05)。与模型组比较,益肾活血丸组大鼠子宫内膜Wnt7a mRNA表达量显著升高(P<0.05),HOXA10、Wnt7a蛋白表达量及PU值显著升高(P<0.05)。相关性分析发现,子宫内膜HOXA10与Wnt7a的表达呈高度正相关。结论:益肾活血丸可通过调控子宫内膜Wnt7a/HOXA10的表达改善子宫内膜容受性。Objectives:To explore the mechanism of Yishen Huoxue Pills regulating Wnt7a/HOXA10 to improve endometrial receptivity.Methods:SPF grade female SD rats were randomly divided into normal group,model group,mifepristone group,Yishen Huoxue pill group,and mifepristone+Yishen Huoxue pill group.The rats in the model group and the Yishen Huoxue Pill group were used to replicate the rat model of poor endometrial receptivity,and the rats in the mifepristone group and the Mifepristone+Yishen Huoxue Pill group were used to replicate the embryo implantation disorder animal model.From the first day of pregnancy,the rats in Yishen Huoxue Pill group and Mifepristone+Yishen Huoxue Pill group were given Yishen Huoxue Pill solution by intragastric administration at a dose of 1.733 g·kg^(-1)·d^(-1),while the other groups were given distilled water by intragastric administration.1 time a day,2 mL each time.The levels of HOXA10 mRNA and Wnt7a mRNA in endometrial tissue were detected by real-time quantitative PCR.The protein levels of HOXA10 and Wnt7a in endometrial tissue were detected by Western blot and immunohistochemical techniques.And correlation analysis was used to investigate the correlation between HOXA10 and Wnt7a.Results:Compared with the normal group,the expression levels of Wnt7a mRNA and HOXA10 mRNA in the endometrium of rats in the model group and mifepristone group were significantly decreased(P<0.05),and the protein expressions and PU values of HOXA10 and Wnt7a were significantly decreased(P<0.05).Compared with the model group,the expression of Wnt7a mRNA in the endometrium of the rats in the Yishen Huoxue Pill group was significantly increased(P<0.05),the protein expression and the PU values of HOXA10,Wnt7a were significantly increased(P<0.05).Correlation analysis found that the expression of endometrial HOXA10 and Wnt7a was highly positively correlated.Conclusion:Yishen Huoxue Pills can improve endometrial receptivity by regulating the expression of endometrial Wnt7a/HOXA10.
