摘 要:目的观察果糖二磷酸钠后处理对老年大鼠体外缺血再灌注心肌的保护作用。方法选择健康清洁级老年雄性SD大鼠60只,随机分为对照组(A组)、缺血再灌注组(B组)、环孢霉素组(C组)和果糖二磷酸钠后处理组(D组),每组15只。记录平衡末(T_(0))、缺血前1 min(T_(1))、再灌注后10 min(T_(2))、20 min(T_(3))和60 min(T_(4))各时间点心率、左心室发展压(LVDP)和左心室压力上升和下降最大速率(±dp/dt_(max))。实验结束后采用电镜观察心肌线粒体变化情况,并检测烟酰胺腺嘌呤二核苷酸(NAD^(+))和线粒体细胞色素C含量。结果B组、C组、D组T_(2)~T_(4)时心率、LVDP和±dp/dt_(max)较A组明显降低,差异有统计学意义(P<0.05);与B组比较,C组、D组T_(2)~T_(4)时心率、LVDP和±dp/dt_(max)明显升高,差异有统计学意义(P<0.05)。与A组比较,B组、D组线粒体表面积与体积比明显降低,差异有统计学意义(P<0.05);与B组比较,C组、D组线粒体表面积与体积比明显升高,差异有统计学意义(P<0.05)。B组线粒体直径较A组明显增加,而C组、D组线粒体直径较B组明显降低,差异有统计学意义(P<0.05)。与A组比较,B组、C组、D组心肌细胞NAD^(+)和线粒体细胞色素C含量明显减少,差异有统计学意义(P<0.05);与B组比较,C组、D组心肌细胞NAD^(+)和线粒体细胞色素C含量明显升高[(9.17±0.96)μmol/L、(8.39±0.57)μmol/L vs(6.68±0.70)μmol/L,(0.37±0.06)μmol/g、(0.34±0.07)μmol/g vs(0.23±0.05)μmol/g,P<0.05]。结论果糖二磷酸钠后处理通过抑制线粒体通透性转换孔开放保护线粒体,减轻缺血再灌注对老年大鼠体外心肌的损伤。Objective To study the role of fructose-sodium diphosphate in protecting myocardium against in vitro ischemia/reperfusion(I/R)injury in aged rats.Methods Sixty healthy aged male SD rats were randomly divided into control group or group A,I/R group or group B,cyclosporine group or group C and postconditioning fructose-sodium diphosphate group or group D(15 in each group).Their heart rate,left ventricular development pressure(LVDP),maximal increase and decrease rates of left ventricular pressure(±dp/dt_(max))were recorded at the end of equilibrium(T_(0)),1 min before ischemia(T_(1)),10 min after reperfusion(T_(2)),20 min(T_(3))after reperfusion and 60 min(T_(4))after reperfusion.The changes of myocardial mitochondria were observed by transmission electron microscopy and the serum levels of NAD^(+) and mitochondrial cytochrome C in different groups were detected at the end of experiment.Results The heart rate,LVDP and±dp/dt_(max) were significantly lower in groups B,C and D at T_(2)-T_(4)(P<0.05)and significantly higher in groups C and D than in group B at T_(2)-T_(4)(P<0.05).The surface area/volume ratio of mitochondria was significantly lower in groups B and D than in group A(P<0.05)and signifi-cantly higher in groups C and D than in group B(P<0.05).The diameter of mitochondria was significantly longer in group B than in group A and significantly shorter in groups C and D than in group B(P<0.05).The serum levels of NAD^(+) and mitochondrial cytochrome C were significant-ly lower in groups B,C and D than in group A(P<0.05)and significantly higher in groups C and D than in group B(9.17±0.96μmol/L and 8.39±0.57μmol/L vs 6.68±0.70μmol/L,0.37±0.06μmol/g and 0.34±0.07μmol/g vs 0.23±0.05μmol/g,P<0.05).Conclusion Postconditioning fructose-sodium diphosphate can alleviate the in vitro myocardial I/R injury by inhibiting the mitochondrial permeability pole opening in aged rats.
